长链非编码RNA牛磺酸上调基因1在糖尿病肾病中的表达及意义

Expression and significance of lncRNA TUG1 in diabetic nephropathy

目的:探究长链非编码RNA牛磺酸上调基因1(lncRNA TUG1)在糖尿病肾病中的表达及意义。方法:选择糖尿病肾病患者65例为观察组,选择同期肾微小病变患者65例为对照组。均留取病变的穿刺活检组织和患者空腹静脉血,应用实时荧光定量PCR检测病变组织和血清中lncRNA TUG1的表达。比较两组患者病变组织和血清lncRNA TUG1表达水平。比较不同临床病理特征糖尿病肾病患者病变组织和血清lncRNA TUG1表达水平。分析观察组患者病变组织与血清lncRNA TUG1表达的相关性。结果:观察组病变组织和血清中lncRNA TUG1表达水平明显低于对照组(均P<0.05)。lncRNA TUG1在不同病变类型、不同硬化肾小球硬化占比患者病变组织和血清中的表达比较差异有统计学意义(均P<0.05),而在不同性别、年龄患者病变组织和血清中的表达比较差异无统计学意义(均P>0.05)。糖尿病肾病患者病变组织与血清lncRNA TUG1表达呈正相关(r=0.693,P=0.021)。结论:糖尿病肾病患者病变组织与血清lncRNA TUG1表达下降且呈正相关,检测血清lncRNA TUG1表达可能对预测病变的形成有一定价值。

Objective:To investigate the expression and significance of long non-coding RNA taurine up-regulated gene 1(lncRNA TUG1)in diabetic nephropathy.Methods:A total of 65 patients with diabetic nephropathy were selected as the observation group,and 65 patients with microscopic renal lesions in the same period were selected as the control group.The lesion biopsy tissues and fasting venous blood of all patients were collected,and the expression of lncRNA TUG1 in the lesion tissue and serum was detected by qRT-PCR.The expression levels of lncRNA TUG1 in lesion tissue and serum were compared between the two groups.The expression levels of lncRNA TUG1 in lesion tissue and serum of diabetic nephropathy patients with different clinicopathological characteristics were compared.The correlation between lesion tissue and serum lncRNA TUG1 expression in the observation group was analyzed.Results:The expression levels of lncRNA TUG1 in the lesion tissue and serum of the observation group were significantly lower than those of the control group(all P<0.05).There were significant differences in the expression of lncRNA TUG1 in lesion tissue and serum of patients with different lesion types and different proportions of sclerotic glomerulosclerosis(all P<0.05),while there were no significant differences in the expression of lncRNA TUG1 in lesion tissue and serum of patients with different gender and age(all P>0.05).There was a positive correlation between the expression of lncRNA TUG1 in lesion tissue and serum in patients with diabetic nephropathy(r=0.693,P=0.021).Conclusion:The expression of lncRNA TUG1 in lesion tissue and serum of patients with diabetic nephropathy is decreased and positively correlated.Detection of serum lncRNA TUG1 expression may have certain value in predicting the formation of diabetic nephropathy.
