摘要
目的:观察筋骨痛消丸对骨关节炎(OA)大鼠关节软骨和巨噬细胞炎症模型中组织生长因子-β_(1)(TGF-β_(1))表达的影响。方法:采用木瓜蛋白酶关节腔注射法,建立SD大鼠骨关节炎模型,60只SD大鼠随机分为对照组、模型组、塞来昔布组及筋骨痛消丸低、中、高剂量治疗组。连续灌胃给药4周,并于4周末处死大鼠,免疫组化检查各组大鼠关节软骨TGF-β_(1)的表达情况。利用LPS诱导RAW264.7细胞炎症模型,并采用筋骨痛消丸进行干预,分别在mRNA及蛋白水平上观察TGF-β_(1)的表达。结果:与对照组相比,OA模型组大鼠TGF-β_(1)表达明显提高(P<0.05);与模型组相比,筋骨痛消丸高剂量组TGF-β_(1)表达明显降低(P<0.05)。在巨噬细胞炎症模型中,与对照组相比,模型组在mRNA及蛋白水平TGF-β_(1)均明显提高(P<0.05);与模型组相比,筋骨痛消丸低、中、高剂量组呈现剂量依赖地降低TGF-β_(1)的mRNA及蛋白表达。结论:筋骨痛消丸能够降低OA大鼠模型关节软骨及巨噬细胞炎症模型中TGF-β_(1)的表达,为进一步探讨其对OA的治疗作用机制提供了依据。
Objective:To observe the effect of Jingu Tongxiao Pill on tissue growth factor-β_(1)(TGF-β_(1)) expression in the articular cartilage of rats with osteoarthritis(OA) and the inflammatory model of macrophages.Methods:Papain intra-articular injection was used to establish the model of SD rat with osteoarthritis.60 SD rats were randomly divided into control group, model group, celecoxib group, Jingu Tongxiao Pill low dose group, medium dose group and high dose group.The rats were intragastrically administered for 4 weeks, and were killed at the end of the 4 th week.The TGF-β_(1)expression of articular cartilage in each group was examined by immunohistochemistry.The inflammation model of RAW264.7 cell was induced by LPS,and intervened with Jingu Tongxiao Pill.TGF-β_(1)expression was observed on both the mRNA and protein level.Results:Compared with control group, the TGF-β_(1)expression of the OA model group rats was increased significantly(P<0.05);Compared with model group, the TGF-β_(1)expression of Jingu Tongxiao Pill high dose group was obviously lower(P<0.05).In macrophage inflammatory models, compared with the control group, the TGF-β_(1)expression of the model group on both the mRNA and protein level were significantly increased(P<0.05);Compared with model group, the group of Jingu Tongxiao Pill with low, medium and high dose was dose-dependent to reduce mRNA and protein expression of TGF-β_(1).Conclusion:Jingu Tongxiao Pill can reduce the expression of TGF-β_(1)in the articular cartilage of rats with OA and the inflammatory model of macrophages, which provides a certain basis for further exploring its therapeutic mechanism on OA.
