摘要
目的:研究细辛挥发油对口腔主要致龋菌变异链球菌和黏性放线菌生长、黏附能力及产胞外多糖的影响,探索细辛挥发油的防龋作用。方法:将细辛挥发油按照二倍梯度稀释法测定最低抑菌浓度MIC和最低杀菌浓度MBC,再以低于MIC 4个浓度梯度配制含药TPY的液体培养基,将0.05%氯己定溶液作为阳性对照组,不含药组作为阴性对照组;采用酶标仪测定吸光度值从而判定细菌黏附能力;通过蒽酮法测定细菌产胞外多糖的情况。结果:细辛挥发油对变异链球菌的MIC为4.375%、MBC为5%;对黏性放线菌的MIC为3.75%、MBC为4.375%;MIC、1/2MIC两个浓度组对主要致龋菌的黏附抑制率、抑制其产胞外多糖效果明显(P<0.05)。结论::MIC、1/2MIC浓度细辛挥发油能够抑制口腔主要致龋菌的生长、黏附及产胞外多糖,其中对变形链球菌的抑制效果更为显著,说明该浓度的细辛挥发油在龋病预防方面有潜在的应用价值。
Objective:To study the effect of Asarum volatile oil on the growth, adhesion ability and exopolysaccharide production of Streptococcus mutans and Actinomyces viscosus, and to explore the mechanism of anti-caries effect of Asarum volatile oil.Methods:The minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of Asarum volatile oil were determined by double gradient dilution method, and then the medicated TPY liquid medium was prepared at four concentrations lower than the MIC.0.05% chlorhexidine solution was used as the positive control, and the negative control group did not contain the liquid;the bacterial adhesion ability was determined by microplate reader;the production of extracellular polysaccharides by bacteria was determined by anthrone method.Results:The MIC of Asarum volatile oil was 4.375% and MBC was 5% for Streptococcus mutans, 3.75% and 4.375% for Actinomyces viscosus, and the adhesion inhibition rate and inhibition effect of exopolysaccharide were obvious in MIC and 1/2 MIC groups(P<0.05).Conclusion:The volatile oil of Asarum at MIC and 1/2 MIC concentrations can inhibit the growth, adhesion and sugar production of the main cariogenic bacteria in the oral cavity, and the inhibitory effect on Streptococcus mutans is more significant, which has a prospect in caries prevention.
