摘要
为探究双酚A(BPA)是否通过调控抗苗勒管激素(AMH)表达量改变而引起青春期雌性小鼠卵巢功能和结构的变化。将24只雌性小鼠(昆明白)随机分为4组,每组6只。3组为实验组,分别给予10 mg/kg、50 mg/kg和250 mg/kg BPA灌胃处理;1组为对照组,灌胃等体积的玉米油。1周后处死各组小鼠,取各组实验动物卵巢制作组织切片,HE染色观察各组形态结构变化,免疫组织方法分析不同组卵巢中AMH表达的变化。结果表明,与对照组比较,10 mg/kg和50 mg/kg BPA剂量组小鼠卵巢组织未见明显异常,卵巢组织结构相对完整。250 mg/kg BPA剂量组小鼠卵巢组织中原始卵泡、初级卵泡、次级卵泡数减少,闭锁卵泡数增加,卵巢组织结构出现明显的病理损伤等改变。免疫组织化学显示,小鼠染毒BPA在250 mg/kg时,小鼠卵巢AMH表达量显著升高,说明高剂量BPA可引起青春期雌性小鼠卵巢结构的改变和组织中AMH蛋白表达升高,通过改变卵巢的储备功能会导致生殖相关疾病。
In order to explore whether bisphenol A(BPA)could induce changes in ovarian function and structure in pubertal female mice by regulating the expression of anti-Müllerian hormone(AMH),24 female mice(Kunbai)are randomly divided into 4 groups with 6 mice in each group.Among them,3 groups are the experimental groups,which are given 10 mg/kg,50 mg/kg and 250 mg/kg BPA by gavage,respectively.And the same volume of corn oil is given by gavage in control group.One week later,the mice in each group are sacrificed,and the ovaries of each group of experimental animals are taken to make tissue sections.The morphological and structural changes of each group are observed by HE staining,and the changes of AMH expression in the ovaries of different groups are analyzed by immunohistochemistry.Compared with the control group,there is no obvious abnormality in the ovarian tissue of the mice in 10 mg/kg and 50 mg/kg BPA dose groups,and the ovarian tissue structure is relatively intact.In the 250 mg/kg BPA dose group,the number of primordial follicles,primary follicles and secondary follicles decrease,the number of atretic follicles increases,and the ovarian tissue structure show obvious pathological damage,etc.Immunohistochemistry shows that the expression of AMH in the ovaries of mice exposes to BPA at 250mg/kg significantly increases.High-dose BPA can cause changes in ovarian structure and increase AMH protein expression in the tissues of adolescent female mice,and lead to reproductive-related diseases by changing the ovarian reserve function.
