变温层积过程中皇后帝王花种子内源激素含量的变化

Changes of Endogenous Hormone Contents in Seeds of Protea magnifica during Warm-cold Stratification Treatment

摘要 皇后帝王花(Protea magnifica)种子具有休眠习性,不经任何处理很难发芽,限制了其在中国的引种试验与推广。本研究采用酶联免疫吸附法测定变温层积(光照10 h温度20℃,黑暗14 h温度10℃)过程中皇后帝王花种子内源激素ABA、GA3、IAA、ZR的含量及其动态变化,来探索其与种子休眠解除的内在联系。在变温层积过程中,随着时间的增加,皇后帝王花种子内ABA含量整体呈下降趋势,在层积第20天时降至低峰68.49 ng/g,比CK减少21.38 ng/g,20 d之后ABA含量变化不大;GA3含量在前15 d缓慢上升,第15天时达到峰值5.53 ng/g而后缓慢下降;IAA含量整体呈先升高后降低的趋势,第25天时陡然达到高峰44.25 ng/g,比CK增加了19.71 ng/g,随后开始下降;ZR含量随着层积时间的增加整体呈缓慢上升趋势,第25天时出现峰值7.63 ng/g,随后开始下降。在变温层积过程中,皇后帝王花种子的GA3/ABA、IAA/ABA、ZR/ABA、(GA3+IAA+ZR)/ABA值逐渐增大,层积第25天时比值最大。变温层积处理能够有效解除皇后帝王花种子的休眠,打破种子内源激素之间的平衡,促进种子的萌发。 The seeds of Protea magnifica are dormant and difficult to germinate without any treatment,which limits its introduction test and popularization in China.In this study,enzyme-linked immunosorbent assay(ELISA)was used to determine the contents and dynamic changes of endogenous hormones ABA,IAA,GA3and ZR in the seeds of P.magnifica during warm-cold stratification treatment(light 10 h,temperature 20℃;dark14 h,temperature 10℃),to explore its internal relationship with the release of seed dormancy.In the process of warm-cold stratification treatment,with the increase of time,the ABA content in the seeds of P.magnifica showed a down ward trend as a whole,and decreased to a low peak 68.49 ng/g on the 20thday of stratification,which was 21.38 ng/g lower than that of CK.The content of ABA changed little after 20 days.The content of GA3increased slowly in the first 15 days,peaked at 5.53 ng/g on the 15thday,and then decreased slowly.The IAA content increased first and then decreased as a whole.On the 25thday,it suddenly reached the peak 44.25 ng/g,increased by 19.71 ng/g compared with CK,and then began to decrease.The content of ZR increased slowly with the increase of stratification time,peaked at the 25thday 7.63 ng/g,and then began to decrease.In the process of warm-cold stratification treatment,the values of GA3/ABA,IAA/ABA,ZR/ABA,(GA3+IAA+ZR)/ABA gradually increased,and the ratio reached the maximum on the 25thday.Warm-cold stratification treatment can effectively release the dormancy of Protea magnifica seeds,break the balance between endogenous hormones,and promote seed germination.
