超声联合微泡对卡铂化疗A549肺癌细胞凋亡影响的实验研究 被引量:1

摘要 目的探讨超声联合微泡在卡铂化疗A549肺癌细胞凋亡中的作用。方法将A549细胞依据不同处理方式分为对照组(未经任何处理)、US组(仅使用超声辐照)、USMB组(使用超声辐照联合微泡)、CBP组(仅使用卡铂)、CBP+US组(使用超声辐照+卡铂)、CBP+USMB组(使用超声辐照联合微泡+卡铂),其中卡铂采用最适宜剂量50μg/ml,超声辐照参数:声强0.6 W/cm^(2),辐照时间60 s。处理后培养24 h,每组分别消化、离心、收集细胞,加入FITC标记的Annexin V、PI室温避光15 min,使用流式细胞术检测各组A549肺癌细胞早期凋亡率、晚期凋亡率、总凋亡率,并对检测结果进行比较。结果A549肺癌细胞总凋亡率由低到高依次为对照组、US组、USMB组、CBP组、CBP+US组、CBP+USMB组,其中CBP+USMB组早期凋亡率、晚期凋亡率、总凋亡率与其余各组比较,差异均有统计学意义(均P<0.01);CBP+US组与CBP组晚期凋亡率、总凋亡率比较,差异均有统计学意义(均P<0.01)。结论超声联合微泡在卡铂化疗A549肺癌细胞凋亡中有协同作用。 Objective To investigate the effect of ultrasound combined with microbubbles on apoptosis of A549 cells treated with carboplatin.Methods A549 cells were divided into control group(without any treatment),US group(ultrasonic irradiation only),USMB group(ultrasonic irradiation combined with microbubbles),CBP group(carboplatin only),CBP+US group(ultrasonic irradiation combined with carboplatin)and CBP+USMB group(ultrasonic irradiation combined with microbubbles and carboplatin)according to different treatment methods,in which carboplatin was used at an optimal dose of 50μg/ml,ultrasonic irradiation was used at the intensity of 0.6 W/cm^(2),and the irradiation time was 60 s.After 24 h of treatment,the cells in each group were detached,centrifuged and collected.FITC-labeled Annexin V and PI were added for 15 min.The early,late and total apoptosis rate of A549 cells were detected by flow cytometry,and the differences were compared.Results The total apoptosis rate of A549 cells from low to high was in the order of control group,US group,USMB group,CBP group,CBP+US group,CBP+USMB group.The early,late and total apoptosis rates of CBP+USMB group were significantly different compared with those of the rest groups(all P<0.01).The late and total apoptosis rates between the CBP+US group and CBP group were significantly different(both P<0.01).Conclusion Ultrasound combined with microbubbles play a synergistic role in carboplatininduced apoptosis of A549 cells.
