涤痰解毒方对有机磷农药中毒模型大鼠线粒体呼吸链复合物的影响 被引量:1

Effects of Ditan Jiedu Prescription on Mitochondrial Respiratory Chain Complexes in Rats with Organophosphorus Pesticide Poisoning

摘要 目的 观察涤痰解毒方(涤痰汤加减)对急性有机磷农药中毒(acute organophosphorus pesticide poisoning,AOPP)模型大鼠肺组织及线粒体呼吸链复合物活性及三磷酸腺苷(ATP)含量的影响,以探讨其治疗作用。方法 SD雄性大鼠灌胃敌敌畏溶液造模,模型制备成功后,随机分为模型组、西药(长托宁)组、西药+等剂量涤痰解毒方组、等剂量涤痰解毒方组、高剂量涤痰解毒方组,分别进行相应干预。治疗结束后取一侧肺组织,石蜡包埋、切片,HE染色观察;取另一侧肺组织,分离肺组织线粒体,采用比色法,按照线粒体呼吸链复合物测定试剂盒说明书进行线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性及三磷酸腺苷(ATP)含量的检测。结果 有机磷处理后的大鼠肺组织损伤明显,西药、西药+涤痰解毒方及等剂量、高剂量涤痰解毒方治疗后肺损伤均明显减轻;有机磷处理后线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性及ATP含量明显降低(P<0.05),等剂量及高剂量涤痰解毒方组均可上调线粒体复合物Ⅰ含量(P<0.05);西药及西药+等剂量涤痰解毒方组均可上调线粒体复合物Ⅱ含量(P<0.05);所有治疗组均可升高线粒体呼吸链复合物Ⅲ、Ⅳ含量(P<0.05)。各治疗组均能明显上调ATP含量(P<0.05)。结论 涤痰解毒方可减轻AOPP中毒模型大鼠肺组织损伤程度,可通过调节线粒体呼吸链复合物活性改善线粒体呼吸功能和能量代谢,发挥治疗作用。 Objective To observe the effect of Ditan Jiedu Prescription(modified Ditan Decoction)on the activity of lung tissue and mitochondrial respiratory chain complex and adenosine triphosphate(ATP) content in model rats induced by acute organophosphorus pesticide poisoning(AOPP),and to explore its therapeutic effect. Methods SD male rats were molded with Dichlorvos Solution. After the models were successfully prepared,they were randomly divided into model group,western medicine(Penehyclidine Hydrochloride)treatment group,western medicine +equal-dose of Ditan Jiedu Prescription group,equal-and high-dose of Ditan Jiedu Prescription groups. The groups were intervened respectively. At the end of treatment,unilateral lung tissue was isolated,embedded in paraffin,sectioned,and subjected to HE staining. In addition,mitochondria separated from another side of lung tissue was observed by colorimetry. The activities of mitochondrial respiratory chain complex(I, II, III and IV) and the content of ATP were detected according to micro mitochondrial respiratory chain complex activity assay kit. Results The damage of lung tissue in rats treated with organophosphorus was obvious,and the damage in lung tissue was obviously alleviated after the treatment of western medicine, western medicine + Ditan Jiedu Prescription group,equal-and high-dose of Ditan Jiedu Prescription groups. The activities of mitochondrial respiratory chain complex(I, II, III and IV)and the content of ATP were significantly decreased(P<0.05)after the treatment of organophosphorus. However,the content of mitochondrial complex I was up-regulated in both the equal-and highdose Ditan Jiedu Prescription groups(P<0.05),while the content of mitochondrial complex II was also increased in western medicine group and western medicine+equal-dose of Ditan Jiedu Prescription group(P<0.05). It was found that the content of mitochondrial respiratory chain complex III and IV was markedly increased in all treatment groups(P<0.05). All treatment groups could significantly up-regulate ATP content(P<0.05). Conclusion Ditan Jiedu Prescription can reduce the degree of lung tissue damage in model rats induced by AOPP. It can improve mitochondrial respiratory function and energy metabolism by regulating the activities of mitochondrial respiratory chain complexes,and thus play a therapeutic role.
