调控鸡黑色素生成相关基因共表达网络的构建 被引量:1

摘要 为了挖掘与鸡黑色素生成有关的基因,研究通过对NCBI网站公布的12份惠阳髯毛鸡羽毛毛囊的转录组数据进行了基因差异表达分析,并结合黑色素生成相关通路中的基因构建了共表达网络。结果显示:共筛选出了74个差异基因和100个通路基因用于权重基因共表达网络分析(Weighted Gene Co-expression Network Analysis,WGCNA),在分析得到的5个模块中,青色模块与羽色表型显著相关(R=0.94,P<0.01)。对该模块进行hub基因挖掘,共获得16个hub基因。对该模块进行GO,KEGG富集分析,发现被显著富集的αMSH/MC1R,WNT/β-catenin信号通路参与了黑色素生成的调控。综合基因差异表达,WGCNA,GO和KEGG多种分析方法,筛选出基因WNT1在黑色素生成中起关键作用。 Gene differential expression analysis was performed on the transcriptome data of 12 feather follicles of Huiyang bearded chicken published on NCBI website to mining genes related to melanin production of chiken,and the co-expression network was constructed by combining the genes in the melanin production related pathway.The results showed that a total of 74 differential genes and 100 pathway genes were screened for WGCNA analysis,among the five modules obtained after analysis,turquoise module was significantly correlated with feather color phenotype(R=0.94,P<0.01).A total of 16 hub genes were obtained by hub gene mining for this module.Go and KEGG enrichment analysis was performed on this module,andαMSH/MC1R,WNT/β-Catenin signaling pathway were significantly enriched and involved in melanin production regulation.Multiple analysis methods of gene differential expression,WGCNA,GO and KEGG were integrated and the gene WNT1 that played a key role in melanin production was screened.
