金雀异黄素对免疫抑制大鼠骨骼肌疲劳的影响

摘要 通过对免疫抑制大鼠进行负重游泳试验研究金雀异黄素缓解骨骼肌疲劳的作用效果。选取72只雄性SD大鼠,按照大鼠体重随机分为空白对照组、免疫抑制模型组、金雀异黄素低剂量组、中剂量组、高剂量组、阳性对照组。除空白对照组外,其余大鼠腹腔注射环磷胺,建立大鼠免疫抑制模型后连续灌胃21 d。通过游泳训练造成疲劳后,测定各组大鼠负重力竭游泳时间、血清中白细胞介素2(IL-2)、干扰素γ(IFN-γ)水平、总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性的变化,骨骼肌中肌酸激酶(CK)、乳酸脱氢酶(LDH)、SOD和GSH-PX活性。结果表明:与免疫抑制模型组相比,金雀异黄素可显著提高大鼠力竭游泳时间,其中高剂量组大鼠力竭游泳时间延长85.76%;高剂量组血清中IL-2含量高达203.36 pg·mL^(-1),较模型组提高了68.33%;血清中IFN-γ含量增至113.24 pg·mL^(-1),较模型组差异极其显著。同时金雀异黄素可以维持运动后大鼠血清和骨骼肌中SOD和GSH-PX活性,其中高剂量组活性显著高于空白对照组;显著提高骨骼肌中CK和LDH含量。由此可见,金雀异黄素可有效提高试验大鼠运动耐力,可以调节血清中免疫指标和骨骼肌中疲劳相关生化指标的水平,对免疫抑制大鼠具有较好的缓解运动性疲劳的效果。 The effect of genistein on relieving fatigue was studied by weight-bearing swimming experiment in immunosuppressed rats.Seventy-two male SD rats were randomly divided into blank control group,immunosuppressive model group,genistein low-dose group,medium-dose group,high-dose group and positive control group.Except the blank control group,the other rats were intraperitoneally injected with cyclophosphamide.And then the immunosuppressive model was established,the rats were continuously gavaged for 21 days.After fatigue caused by swimming training,the time of exhaustive swimming with load,the levels of interleukin-2(IL-2),interferon-γ(IFN-γ),the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)in each group were measured.Activity of creatine kinase(CK),lactate dehydrogenase(LDH),SOD and GSH-PX in skeletal muscle.The results showed that,compared with immunosuppressive model group,genistein could significantly increase the exhaustive swimming time of rats,in which the exhaustive swimming time of rats in high-dose group was prolonged by 85.76%.The content of IL-2 in serum of high-dose group was as high as 203.36 pg·mL^(-1),which was 68.33%higher than that of model group.The serum IFN-γcontent increased to 113.24 pg·mL^(-1),which was significantly different from that of the model group.At the same time,it can maintain the activity of SOD and GSH-PX in rats after exercise,in which the activity of high-dose group is significantly higher than that of blank control group.Significantly increase the contents of CK and LDH in skeletal muscle.Therefore,genistein can effectively improve the exercise endurance of experimental rats,adjust the levels of immune indexes in serum and fatigue-related biochemical indexes in skeletal muscle,and has a good effect on relieving exercise-induced fatigue in immunosuppressed rats.
