猪肉中雄烯酮含量不同对小鼠性腺组织及性激素水平的影响

Effects of Different Contents of Androstenone in Pork on Gonadal Tissue in Mice

摘要 探究小鼠摄入雄烯酮(ADT)含量不同的猪肉后,其性腺组织及性激素水平的变化。采用Elisa法检测不同来源猪肉中雄烯酮含量高低。60只KM雄性小鼠与60只KM雌性小鼠随机分为5组,分别为空白对照组、低雄烯酮含量猪肉喂养组(两供试量组分别为5和10 g/kg)、高雄烯酮含量猪肉喂养组(两供试量组分别为5和10 g/kg),雌雄各半;连续喂养8周。试验结束后,动物深度麻醉后取血,采用安乐死并解剖,取关键脏器。计算小鼠前列腺及睾丸指数、卵巢指数,对组织进行苏木精-伊红染色。检测雄性小鼠血清中睾酮及雌性小鼠血清中雌二醇含量。结果表明,与空白组相比,高雄烯酮含量的猪肉喂食后雄性小鼠出现一定程度的躁狂状态,动物之间撕咬情况严重,血清睾酮水平显著增加,并呈剂量依赖性;低雄烯酮猪肉喂养组撕咬情况不显著,睾酮水平未见显著升高;高雄烯酮猪肉喂养后雌性小鼠血清雌二醇含量显著性降低,呈剂量依赖性;猪肉喂养组动物各组织器官病变程度不显著,雄烯酮含量偏高的猪肉摄入一段时间后可能将引起雄性小鼠睾酮水平升高、雌性小鼠雌二醇水平下降,但未发现明显引起动物性组织器官病变。 To investigate the effects of pork containing different level of androstenone(ADT) on gonadal tissue in mice, the content of androstenone in pork from different sources was determined by Elisa. The 60 KM male mice and 60 KM female mice were randomly divided into 5 groups, including bank control group, low-ADT-content pork feeding groups(two groups with 5 and 10 g/kg respectively), high-ADT-content pork feeding groups(two groups with 5 and 10 g/kg respectively), with 12 male and 12 female mice in each group. All the groups were fed with certain amount of pork for eight weeks except bank control group. The levels of testosterone in serum of male mice and estradiol in serum of female mice were measured at the end of study. The prostate and testicular indexes of male mice and ovary indexes of female mice were calculated.haematoxylin-eosin staining was performed on the above tissues. Results showed that compared with the bank control group,male mice fed with high-ADT-content pork showed much more aggressive accompanied by biting and maniacal behavior with a dose-dependence manner. No obvious abnormal behaviors were showed in control and low-ADT-content pork treating groups. Serum level of testosterone increased significantly in the high-ADT-content pork giving group compared to blank group. The serum estradiol content of female mice significantly decreased in the high-ADT-content pork giving group compared to blank group. No marked pathological changes were found in each group. Treating high-ADT-content pork might influence the sex hormone levels in mice.
