双水相分离技术应用于蛋白质纯化的研究进展

Research Progress of Aqueous Two-Phase System in Protein Purification

摘要 双水相系统以其操作简单、成本低廉和强大的可调节性在众多纯化方法中脱颖而出。综述双水相分离技术的发展历史、技术原理及应用范围,重点对提取方法进行分类与总结。简要分析传统双水相、亲和双水相、离子液体双水相、双水相逆流色谱的原理,总结不同双水相体系在蛋白纯化应用中的最新研究进展。 Aqueous two-phase system(ATPS) stands out among many purification methods because of its simple operation,low cost and strong controllability. The development history, technical principle and application range of aqueous two phase separation technology are summarized, with emphasis on the classification and summary of extraction methods. The principles of traditional aqueous two-phase systems, affinity-based aqueous two-phase systems, ionic liquid two-phase and aqueous two-phase countercurrent chromatography are briefly analyzed, and the latest research progress in the application of different aqueous two-phase systems in protein purification is summarized.
