加味四妙勇安膏对急性放射性皮炎大鼠皮肤细胞凋亡和炎症损伤的影响 被引量:4

Effects of Modified Simiao Yongan Ointment on apoptosis of wound skin and inflammatory injury in rats with acute radiation dermatitis

摘要 目的:观察加味四妙勇安膏对急性放射性皮炎(a-RISR)大鼠创面愈合部位细胞凋亡及炎症损伤的影响。方法:18只SPF级雄性Wistar大鼠随机分为空白对照组(Control组)、模型组(Model组)、加味四妙勇安膏A组(A7、A14组)和加味四妙勇安膏B组(B7、B14组),每组3只。除Control组,其他组均建立Ⅱ度a-RISR大鼠,分别用加味四妙勇安膏或0.9%氯化钠溶液对照射野进行外涂,观察照射野皮肤组织淋巴细胞浸润、细胞凋亡及线粒体损伤情况,检测血清白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)水平。结果:与Control组比较,Model组大鼠皮损组织可见大量淋巴细胞浸润,凋亡细胞数目较多,线粒体空泡化异常伴有数量减少,血清IL-1β、IL-6、TNF-α水平显著升高(P<0.01)。与Model组比较,A组和B组明显改善照射野皮肤淋巴细胞浸润,减少创面凋亡细胞数量,缓解线粒体损伤,显著降低大鼠血清IL-1β、IL-6、TNF-α水平(P<0.01,P<0.05)。结论:加味四妙勇安膏外用可通过调节照射野皮肤细胞凋亡和炎症损伤达到促进a-RISR大鼠创面愈合的防治作用。 Objective: To observe the effects of Modified Simiao Yongan Ointment on apoptosis of wound healing area and inflammatory injury in rats with acute radiation dermatitis. Methods: Eighteen SPF male Wistar rats were randomly divided into Control group,Model group,Modified Simiao Yongan Ointment A group(A7,A14group) and Modified Simiao Yongan Ointment B group(B7,B14 group),3 rats in each group. Except the Control group,the other groups were established into the rat model of II degree acute radiation dermatitis. Their irradiated field was respectively coated with Modified Simiao Yongan Ointment or 0.9% sodium chloride solution. The lymphocyte infiltration,cell apoptosis and mitochondrial damage of skin tissue in the irradiation field were observed,and the levels of IL-1β,IL-6 and TNF-α in serum were detected. Results: Compared with the Control group,a large number of lymphocytes were infiltrated in the skin tissue of Model group,the number of apoptotic cells was more,mitochondria showed vacuolated and decreased in numbers,and the levels of IL-1β,IL-6 and TNF-α in serum were significantly increased(P<0.01). Compared with Model group,external application of Modified Simiao Yongan Ointment A and B group significantly improved the infiltration of skin lymphocytes in irradiated field,reduced the number of apoptotic cells on wound surface,alleviated mitochondrial damage,and significantly decreased the levels of IL-1β,IL-6 and TNF-α in serum of rats(P<0.01,P<0.05). Conclusion: The external application of Modified Simiao Yongan Ointment can promote the wound healing of a-RISR rats by regulating the apoptosis and inflammatory injury of skin cells in irradiation field.
