基于16S rRNA测序研究小陷胸汤对动脉粥样硬化小鼠肠道菌群结构的影响 被引量:1

Effects of Xiaoxianxiong Decoction on intestinal flora structure of atherosclerosis mice based on 16S rRNA sequencing

摘要 目的:观察小陷胸汤对动脉粥样硬化小鼠肠道菌群的影响。方法:将30只雄性ApoE-/-小鼠随机分为空白对照组(n=6)和造模组(n=24)。喂养8周后,将造模组小鼠随机分为模型组、阿托伐他汀组、小陷胸汤原方组、小陷胸汤方剂组,每组6只。空白对照组与模型组小鼠给予等量无菌0.9%氯化钠溶液,阿托伐他汀组小鼠给药剂量为2.6 mg·kg^(-1)·d^(-1),小陷胸汤原方组给药剂量为16.5 g·kg^(-1)·d^(-1),小陷胸汤方剂组给药剂量为5 g·kg^(-1)·d^(-1)。连续给药8周后取材。用油红O染色、HE染色检测主动脉根部斑块含量,基于16S rRNA基因测序技术分析小鼠肠道菌群。结果:与模型组比较,小陷胸汤原方组、方剂组主动脉根部斑块减少(P<0.01);小陷胸汤可以正向调节AS小鼠肠道菌群的2种差异菌门(拟杆菌门,厚壁菌门)与8种差异菌属(Alistipes属、毛螺菌科-NK4A136-group、拟普雷沃菌属、拟杆菌属、Muribaculum属、罗氏菌属、瘤胃球菌科UCG组014属、普雷沃氏科UCG组001属)。结论:小陷胸汤对AS小鼠具有防治作用,该作用可能改善AS小鼠肠道菌群失调来实现的。 Objective:To observe the effects of Xiaoxianxiong Decoction on intestinal flora of atherosclerosis (AS)mice.Methods:Thirty male ApoE-/-mice were randomly divided into a blank control group (n=6) and a modeling group (n=24).After 8 weeks of feeding,the modeling group were randomly divided into a model group,atorvastatin group,Xiaoxianxiong Decoction original formula group and Xiaoxianxiong Decoction formula group,with 6 rats in each group.The blank control group and model group were given the same amount of sterile 0.9% sodium chloride solution.The dose of atorvastatin group mice was 2.6 mg·kg^(-1)·d^(-1),the dose of Xiaoxianxiong Decoction original group was 16.5 g·kg^(-1)·d^(-1),and the dose of Xiaoxianxiong Decoction group was 5 g·kg^(-1)·d^(-1).The materials were drawn after 8 weeks of continuous administration.The plaque content of aortic root was detected by oil red O staining and HE staining,and the intestinal flora of mice was analyzed based on 16S rRNA gene sequencing technology.Results:Compared with the model group,the plaque of aortic root in Xiaoxianxiong Decoction group and formula group decreased;Xiaoxianxiong Decoction can positively regulate 2 different bacteria (Bacteroidetes,Firmicutes) and 8 different bacteria (Alistipes,Lachnospiraceae_NK4A136 group,Alloprevotella,Bacteroides,Roseburia,Muribaculum,Ruminococcaceae_UCG-014,Prevotellaceae_UCG-001) in the intestinal flora of AS mice.Conclusion:Xiaoxianxiong Decoction has a preventive and therapeutic effect on AS mice,which may be achieved by improving the imbalance of intestinal flora in AS mice.
