5株灵芝菌株母种培养特性研究

A Study on Growth Characteristics of Five Strains of Ganoderma lucidum

摘要 以引进的5株灵芝菌株(HZ、RBZ、HLZ、ZHH、MLZ)为研究对象,比较其在马铃薯葡萄糖琼脂(PDA)及改良后的PDA培养基上的生长特性。结果表明,5株灵芝菌株的菌落直径随培养时间的增长而增大,但不同菌株间差异显著。从培养48 h到满皿前,菌株MLZ的生长速率明显大于其他品种;菌株MLZ和ZHH菌丝洁白且生长速度较快,在PDA培养基上培养6 d内就可长满整个培养皿,菌丝长势明显优于HZ、RBZ和HLZ;在PDA培养基中加入枸杞枝条粉能显著改善菌株HZ、RBZ、HLZ和ZHH菌丝生长状况,但最适于菌株MLZ菌丝生长的培养基为PDA+玉米芯粉。研究结果可为进一步提高灵芝母种质量和促进宁夏灵芝产业发展提供技术支撑。 Taking five strains of Ganoderma lucidum as the subject, the growth characteristics on potato glucose agar(PDA) and modified PDA culture medium were compared. The results showed that the colony diameter of 5 Ganoderma strains improved with the increase of culture time, but the difference among different strains was significantly. From 48 h after inoculations to the strains colonized all the petri dishes, the growth rate of MLZ was obviously higher than that of other strains, and the hyphae of strains MLZ and ZHH were white and grew quickly, the whole culture plate could be grown on PDA medium within 6 days, and the growth of hyphae was obviously superior to HZ, RBZ and HLZ. The mycelial growth of HZ, RBZ, HLZ and ZHH could be significantly improved by adding Lycium barbarum branch powder into PDA medium, but the optimum medium for mycelial growth of MLZ was PDA + corncob powder. The research results can provide technical support for further improving the quality of Ganoderma seed and promoting the development of Ganoderma industry in Ningxia district.
