基于UPLC-QTOF/MS的关黄母颗粒主要化学成分及脑组织移行成分研究 被引量:3

摘要 目的 运用UPLC-QTOF/MS法对关黄母颗粒中化学成分及脑组织移行成分进行分析和鉴定。方法 基于UPLC-QTOF/MS,在0.1%甲酸水-乙腈流动相体系下,Acquity UPLC HSS T3色谱柱色谱分离后进行正负离子扫描,结合文献、数据库检索等方式,对制剂主要化学成分及入脑成分进行分析。结果 共分离鉴定出159种化学成分,包括生物碱类、萜类、黄酮类、皂苷类等多种化合物,并成功鉴定出15种脑组织移行成分,结果显示其主要入脑成分为生物碱及萜类物质。结论 该方法准确稳定,适用于关黄母颗粒中化学成分分析,也可为寻找出该类药物质量标志物及抗抑郁作用物质基础提供数据支撑。 Objective To identify the chemical components and brain tissue transition components in Guanhuangmu granules with a UPLC-QTOF/MS method. Methods Based on UPLC-QTOF/MS equipment, and 0.1% formic acid water-acetonitrile mobile phase system, after chromatographic separation on Acquity UPLC HSS T3 column, both positive and negative ion scannings were performed. The main chemical components and brain tissue migration components of the preparation were identified by literature, database retrieval and other methods. Results Totally 159 chemical components were isolated, including alkaloids, terpenoids, flavonoids, saponins and other compounds,and 15 brain tissue transition components were successfully identified. The main components entering the brain were alkaloids and terpenoids. Conclusion The method is accurate and stable, suitable for the analysis of chemical components in Guanhuangmu granules, which provides data support for determining out the quality markers of this kind of drugs and the material basis of antidepression effect.
