摘要 目的 优选与基准样品指标性成分含量、出膏率和指纹图谱相似度均接近的散偏汤提取工艺。方法 以阿魏酸、芥子碱硫氰酸盐、甘草苷及甘草酸的含量和出膏率作为综合评分的指标,在单因素试验结果的基础上采用Box-Behnken设计-响应面法优化提取工艺。结果优化后的散偏汤提取工艺为加水20倍量、提取60 min、提取1次,全方在此提取工艺条件下所得阿魏酸、芥子碱硫氰酸盐、甘草苷和甘草酸含量分别为40.54 mg、70.87 mg、22.30mg、40.63 mg,出膏率为23.83%,与基准样品的质控标准最为接近。结论 散偏汤提取工艺经Box-Behnken设计-响应面法优化后更加可靠,且符合基准样品质量标准范围,可为后续剂型开发提供参考。 Objective To optimize the extraction process of Sanpian decoction, which is similar to the benchmark sample in indicative component content, extract yield and fingerprint similarity. Methods The content and extract yields of ferulic acid, sinapine thiocyanate, liquiritin and glycyrrhizic acid were taken as the comprehensive scoring indicators. Based on the single factor test, the extraction was optimized by Box-Behnken design-response surface methodology. Results The optimized extraction process of Sanpian decoction included 20 times of water, 60 min of extraction, and extraction once. The content of ferulic acid, sinapine thiocyanate, liquiritin, and glycyrrhizic acid in the whole prescription was 40.54 mg, 70.87 mg, 22.30 mg, and 40.63 mg, respectively. The extract yield was 23.83%, which was closest to the quality control standards of the benchmark sample. Conclusion The extraction process of Sanpian decoction is more reliable after optimization by Box-Behnken design-response surface methodology, and meets the quality standards of the benchmark sample, which can provide reference for the development of subsequent dosage forms.
