基于高效液相色谱法测定不同产地、不同抽芯程度远志药材中4种成分的含量

HPLC determination of 4 components in Polygala tenuifolia from different habitats with different core pulling degrees

目的 建立HPLC法测定不同产地、不同抽芯程度远志药材的主要成分,并比较其含量差异。方法 采用HPLC法对远志药材中西伯利亚远志糖A_(5)、西伯利亚远志糖A_(6)、远志酮、3,6’-二芥子酰基蔗糖4种化学成分的含量进行测定,采用聚类分析法比较其含量差异。结果远志4种成分在320 nm下分离度良好,西伯利亚远志糖A_(5)、西伯利亚远志糖A_(6)、远志酮、3,6’-二芥子酰基蔗糖低、中、高浓度的平均回收率在94.8%~106.8%。成分含量分析结果表明,不同产地的远志药材在不同抽芯程度下4种成分含量差异具统计学意义(P<0.05)。结论 该方法科学、简便、准确,可用于不同产地、不同抽芯程度远志药材成分含量测定。

Objective To establish an HPLC method to determine 4 main components of Polygala tenuifolia from different producing areas with different core pulling degrees, and to compare the content difference. Methods The content of sibiricose A_(5), sibiricose A_(6), Polygala tenuifolia ketone and 3, 6’-disinapoyl sucrose in Polygala tenuifolia was determined by HPLC. The content difference was analyzed by cluster analysis. Results The 4 components in Polygala tenuifolia were well separated at 320 nm. The average recoveries of low, medium and high concentrations of sibiricose A_(5), sibiricose A_(6), polygala tenuifolia ketone and 3, 6’-disinapoyl sucrose ranged from 94.8% to 106.8%. The content of the 4 components of Polygala tenuifolia from different areas was significantly different with different core pulling degrees(P<0.05). Conclusion The method is scientific, simple and accurate. It can be used to determine of the content of Polygala tenuifolia medicinal materials from different habitats with different degrees of core extraction.
