摘要
目的建立卵巢癌荷瘤裸鼠模型并探讨橄榄苦苷对卵巢癌荷瘤裸鼠血清代谢的影响。方法采用动物模型结合液相色谱-质谱(UPLC/Q-TOF MS)与化学计量学模式识别方法,鉴定卵巢癌可能的差异代谢物,研究橄榄苦苷对卵巢癌的治疗作用。结果与生理盐水组相比,橄榄苦苷组的假尿嘧啶核苷显著降低,精氨酸和DHA显著升高,差异有统计学意义。结论橄榄苦苷可通过影响精氨酸代谢途径来抑制肿瘤,假尿嘧啶核苷可能是卵巢癌诊断和预后的潜在生物标志物,DHA可能是橄榄苦苷发挥抗肿瘤作用的重要成分。
Objective To establish an ovarian neoplasms tumor-bearing nude mouse model and explore the effect of oleuropein on serum metabolism of ovarian neoplasms-bearing nude mice.Methods Animal models combined with liquid chromatography-mass spectrometry(UPLC/Q-TOF MS)and chemometric pattern recognition were used to identify possible differential metabolites of ovarian cancer,and to study the therapeutic effect of oleuropein on ovarian neoplasms.Results Compared with the saline group,the pseudouridine in the oleuropein group were significantly reduced,and the arginine and DHA were significantly increased,the difference was statistically significant.Conclusion Oleuropein can inhibit tumors by affecting arginine metabolism,improve intestinal flora;pseudouridine may be a potential biomarker for the diagnosis and prognosis of ovarian neoplasms,DHA may be important components of oleuropein to exert antitumor effect.
