MiR-124-3p和SEPT9与宫颈癌患者临床表现的相关性分析

Correlation analysis of miR-124-3p and SEPT9 with clinical manifestations of cervical cancer patients

目的探讨miR-124-3p和SEPT9在宫颈癌中的表达并分析二者与宫颈癌患者临床表现的相关性。方法收集2016年5月至2019年5月在石家庄市妇幼保健院接受手术切除的患者中获得94份宫颈癌组织样本和匹配的癌旁正常组织样本。qRT-PCR用于检测miR-124-3p表达;免疫组织化学用于分析SEPT9的阳性表达;比较宫颈癌中miR-124-3p和SEPT9表达与临床特征表现的相关性;Strabase网站预测miR-124-3p和SEPT9靶向关系,Spearman等级分析宫颈癌中miR-124-3p和SEPT9表达的相关性;Kaplan-Meier生存分析和COX回归分析探讨miR-124-3p和SEPT9表达与宫颈癌患者预后的关系。结果与癌旁正常组织比较,宫颈癌组织中miR-124-3p相对表达水平水平降低,SEPT9阳性率升高(P<0.05)。宫颈癌组织中miR-124-3p、SEPT9表达均与肿瘤大小、FIGO分期和淋巴结转移相关(P<0.05)。miR-124-3p和SEPT9存在互补作用位点,且在宫颈癌组织中呈负相关(P<0.05)。宫颈癌组织中miR-124-3p低表达患者3年总生存率(29.79%)低于miR-124-3p高表达患者3年总生存率(59.57%),SEPT9阳性表达患者3年总生存率(56.25%)低于SEPT9阴性表达患者3年总生存率(20.00%)(P<0.05)。SEPT9表达、肿瘤大小、FIGO分期、淋巴结转移是宫颈癌预后的独立危险因素(P<0.05)。结论宫颈癌组织中miR-124-3p表达下调,是宫颈癌预后的独立保护因素;SEPT9表达上调,是宫颈癌预后的独立危险因素,二者呈负相关。

Objective To explore the expressions of miR-124-3p and SEPT9 and analyze their correlation with the clinical manifestations of cervical cancer patients.Methods A total of 94 cervical cancer tissue samples and matched adjacent normal tissue samples were collected from patients undergoing surgical resection in Shijiazhuang Maternity&Child Healthcare Hospital from May 2016 to May 2019.QRT-PCR was used to detect miR-124-3p expression,immunohistochemistry was used to analyze the positive expression of SEPT9.The correlation between the expression of miR-124-3p and SEPT9 in cervical cancer and the clinical features were compared.The relationship between miR-124-3p and SEPT9 targeting was predicted by the Strabase website.Spearman rank analysis of the correlation of miR-124-3p and SEPT9 expression in cervical cancer,and Kaplan-Meier survival analysis and COX regression analysis to explore the relationship between miR-124-3p and SEPT9 expression and prognosis of cervical cancer patients.Results Compared with the adjacent normal tissues,the relative expression level of miR-124-3p in cervical cancer tissues was decreased,and the positive rate of SEPT9 was increased(P<0.05).The expressions of miR-124-3p and SEPT9 in cervical cancer tissues were correlated with tumor size,FIGO stage and lymph node metastasis(P<0.05).MiR-124-3p and SEPT9 have complementary action sites,and they are negatively correlated in cervical cancer tissues(P<0.05).The 3-year overall survival rate of patients with low expression of miR-124-3p in cervical cancer tissue(29.79%)was lower than that of patients with high expression of miR-124-3p(59.57%),the 3-year overall survival rate of patients with positive SEPT9 expression(56.25%)was lower than that of patients with negative SEPT9expression(20.00%)(P<0.05).The expression of SEPT9,tumor size,FIGO stage,and lymph node metastasis were independent risk factors for the prognosis of cervical cancer(P<0.05).Conclusion The down-regulated expression of miR-124-3p in cervical cancer tissue is an independent protective factor for the prognosis of cervical cancer;the up-regulated expression of SEPT9 is an independent risk factor for the prognosis of cervical cancer,and the two are negatively correlated.
