从TLR4/NF-κB信号轴探讨小儿解表口服液抗流感病毒药效机理 被引量:3

Based on TLR4/NF-κB signal axis to explore the pharmacodynamic mechanism of Xiaoer jiebiao oral liquid against influenza virus

摘要 目的通过流感病毒滴鼻方法建立甲型H1N1流感病毒模型,从TLR4/NF-κB介导的信号轴探讨小儿解表口服液的治疗作用与其药效机制。方法流感病毒滴鼻方法建立模型,经给药治疗后,计算各组小鼠肺指数;HE染色观察肺部组织病理改变;ELISA法检测血清中IL-1β、TNF-α、IL-6、INF-γ含量;Western blot法检测TLR4、NF-κB p65蛋白情况。结果小儿解表口服液可以较好降低小鼠的肺指数,减轻肺组织损伤;减少IL-1β、IL-6、TNF-α、INF-γ的含量;降低TLR4、NF-κB p65蛋白水平(P<0.05或P<0.01)。结论小儿解表口服液能有效减轻流感病毒感染后小鼠肺部炎性病变,发生作用的重要途径可能是抑制TLR4/NF-κB信号轴的活化从而发挥减轻炎症的作用。 Objective To establish influenza A(H1N1) virus model by influenza virus nasal drip method,from TLR4/NF-κB-mediated signal axis to explore the therapeutic effect and pharmacodynamic mechanism of Xiaoer jiebiao oral liquid.Methods The model was established by influenza virus nasal drip.After administration and treatment,the lung index of mice in each group was calculated;HE staining was used to observe the pathological changes of lung tissue;Detection of IL-1 in serum by ELISA β、 TNF-α、 IL-6、INF-γ Content;TLR4 and NF were detected by Western blot-κB p65 protein.Results Xiaoer jiebiao oral liquid can reduce the lung index and lung tissue injury in mice;Reduce IL-1 β、 IL-6、TNF-α、 INF-γ Content of;Reduce TLR4 and NF-κB p65 protein level(P < 0.05 or P < 0.01).Conclusion Xiaoer jiebiao oral liquid can effectively reduce the pulmonary inflammatory lesions in mice after influenza virus infection.The important way of action may be to inhibit TLR4/NF-κB,The activation of signal axis plays a role in reducing inflammation.
