脾虚症并发白念珠菌感染小鼠血常规及血浆细胞因子的变化及秋泻灵颗粒治疗对其影响 被引量:3

Changes of blood routine and plasma cytokines in mice with spleen deficiency complicated with Candida albicans infection and the effect of Qiuxieling granule treatment on them

摘要 目的 通过检测小鼠血常规、血液生化指标以及血浆中细胞因子含量的变化,探讨脾虚造模并发白念珠菌感染对小鼠免疫功能的影响以及秋泻灵颗粒对其的疗效。方法 72只SPF级昆明小鼠随机分成6组,按照复合因素法制备脾虚模型,脾虚造模结束第2天,脾虚感染模型组和给药治疗各组小鼠经口灌胃白念珠菌悬液(2.5×10~8CFU/mL,0.5mL/只),次日开始按分组给药,每日1次,连续7天。记录各组小鼠实验期间体质量变化,末次给药24h后,处死小鼠,检测血液、肾脏和粪便菌落数,检测免疫器官脏器系数,全血检测血常规,血浆检测尿素氮、肌酐、谷丙转氨酶、谷草转氨酶及TNF-α、IL-12、CCL2、IL-17A、IFN-γ细胞因子含量。结果 脾虚小鼠经口灌胃白念珠菌感染7天后,体重,脾脏、胸腺系数,外周血中白细胞数目、淋巴细胞数目、单核细胞数目、中性粒细胞数目,均显著低于其他各组,血浆中谷丙转氨酶、谷草转氨酶、肌酐的含量均显著高于正常对照组,秋泻灵颗粒给药治疗后可有效降低白念珠菌感染脾虚小鼠的粪便载菌量,显著提升脾脏、胸腺脏器系数,显著提升外周血中白细胞、淋巴细胞、单核细胞和中性粒细胞的数目,上调血浆中TNF-α的水平,降低血浆中谷丙转氨酶、谷草转氨酶和肌酐的含量。结论 脾虚造模可降低小鼠免疫功能,秋泻灵颗粒给药能明显改善小鼠脾虚状态,能有效提高脾虚症并发白念珠菌感染小鼠的免疫功能。 Objective To investigate the effect of spleen deficiency modeling complicated with Candida albicans infection on the immune function of mice and the curative effect of Qiuxieling granules on the immune function of mice by detecting the changes of blood routine,blood biochemical indexes and plasma cytokine content.Methods 72 SPF Kunming mice were randomly divided into 6 groups,and the spleen deficiency model was prepared by compound factor method.On the second day after the establishment of spleen deficiency model,mice in the spleen deficiency infection model group and the treatment group were intragastric with Candida albicans suspension(2.5×108CFU/mL,0.5mL/mouse),and the next day they were given drug treatment by group,once a day for 7 days.The changes of body weight in each group were recorded during the experiment.24h after the last administration,the mice were killed,the number of bacteria in blood,kidney and feces,the coefficient of immune organs were detected,and the blood routine was detected in whole blood.Serum urea nitrogen,creatinine,glutamic-pyruvate transaminase,glutamic-oxalacetic transaminase,TNF-α,IL-12,CCL2,IL-17A,IFN-γ levels were detected.Results After 7 days of oral gavage of Candida albicans infection in spleen deficiency mice,the body weight,spleen and thymus coefficient,the number of leukocytes,lymphocytes,monocytes and neutrophils in peripheral blood were significantly lower than those in other groups,and the contents of alanine aminotransferase,aspartate aminotransferase and creatinine in plasma were significantly higher than those in normal control group,Qiuxieling granule can effectively reduce the fecal bacterial load of Candida albicans infected spleen deficiency mice,significantly improve the organ coefficient of spleen and thymus,significantly increase the number of leukocytes,lymphocytes,monocytes and neutrophils in peripheral blood,and up regulate TNF in plasma-α The levels of alanine aminotransferase,aspartate aminotransferase and creatinine in plasma were decreased.Conclusion The spleen deficiency modeling can reduce the immune function of mice,Qiuxieling granules can obviously improve the state of spleen deficiency in mice,and can effectively improve the immune function of mice with spleen deficiency complicated by Candida albicans infection.
