摘要
通过指纹图谱联合多元统计分析,确定可鉴别土茯苓饮片质量的指标成分。高效液相色谱法(HPLC)被用于15批土茯苓饮片指纹图谱的建立,并采用聚类分析(HCA)、主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)筛选15批土茯苓饮片的差异性指标成分。结果显示,15批土茯苓样品指纹图谱具有9个共有峰,各批样品与对照图谱的相似度均在0.919以上,指纹图谱方法重复性良好;HCA结果表明15批土茯苓样品可被分成3类;PCA结果表明3个主成分能够较好地反映15批土茯苓样品的主要信息;OPLS-DA处理中变量重要性投影(VIP)筛选出的差异性指标成分依VIP值大小分别为色谱峰9号、2号、8号、1号、3号。当前市售土茯苓饮片质量不均,需划分质量等级;土茯苓饮片质控标准有待完善,需进行进一步研究。
The fingerprint of Smilacis Glabrae Rhizoma pieces was established and combined with multivariate statistical analysis method to find and identify the quality differences in Smilacis Glabrae Rhizoma pieces. High perfor-mance liquid chromatography(HPLC) was used to establish the fingerprints of 15 batches of Smilacis Glabrae Rhizoma pieces. Combined with cluster analysis(HCA), principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA), 15 batches were determined to screen out the differential compounds of Smilacis Glabrae Rhizoma pieces. There were 9 common peaks in the fingerprints of the 15 batches of Smilacis Glabrae Rhizoma pieces, and the similarity between each batch of samples and the control spectrum was above 0.919. The fingerprint method had good repeatability. HCA results showed that the 15 batches of Smilacis Glabrae Rhizoma pieces could be divided into three categories;PCA results showed that the three principal components could well reflect the main information of the 15 batches of Smilacis Glabrae Rhizoma pieces;the differential markers screened by variable importance projection(VIP) in the OPLS-DA process were in the order of VIP value: chromatographic peak No.9, No.2, No.8, No.1, No.3. At present, the quality of raw Smilacis Glabrae Rhizoma decoction pieces on the market is uneven and can be divided into quality grades;the quality control standards of Smilacis Glabrae Rhizoma decoction pieces need to be improved, and further research is needed.
