搭载右旋布洛芬缓释型微针的制备及性能测试

摘要 载药缓释型微针可以无痛、有效地释放疫苗激素类药物。以交联透明质酸为基材,分别使用预载药和后载药的方法制备右旋布洛芬缓释型微针,研究载药缓释型微针的最佳制备工艺。对这2种载药方法制备的微针阵列进行了力学性能测试,同时通过体外透皮扩散实验评价了微针的释药性能。实验结果表明,预载药法更适合制备载药缓释型微针,微针阵列的成型较好,并且具有足够的力学性能刺破皮肤。体外透皮扩散实验显示,预载药法制备的右旋布洛芬缓释型微针的释药性能更加优异。微针在6 h时释放了约42%的药物,最终,累计释放了约95%的药物。实验结果证明了载药缓释型微针的有效性,为疫苗、激素类药物的临床应用提供了新的研究方向。 Vaccine hormone drugs could be released painlessly and effectively by slow-release microneedles.In order to explore the best preparation process of drug-loaded sustained-release microneedles,cross-linked hyaluronic acid was used as the base material to prepare dextroibuprofen sustained-release microneedles using pre-loading and post-drug loading methods.The mechanical properties of the microneedle arrays prepared by these two drug loading methods were tested,and the drug release performance of the microneedles was evaluated through in vitro transdermal diffusion experiments.The experimental results showed that the preloaded drug method was more suitable for preparing drug-loaded sustained-release microneedles.The microneedle array was well formed and possesses sufficient mechanical properties to pierce the skin.In vitro transdermal diffusion experiments showed that the dexibuprofen sustained-release microneedles prepared by the preloading method had better drug release performance.The microneedle released about 42%of the drug at 6 h,and finally released about 95%of the drug.The experimental results verified the effectiveness of drug-loaded sustained-release microneedles,and developed a new approach for the clinical application of vaccines and hormone drugs.
