刺糖多糖对鸡免疫新城疫疫苗后免疫功能的调节作用 被引量:1

摘要 为了研究刺糖多糖(AH)对鸡免疫功能和新城疫抗体效价的影响,选取150只1日龄海兰褐公鸡,随机分为5组,每组30只,待母源抗体低至2.23log2时,除空白对照组外,其余各组都进行新城疫疫苗免疫,空白对照组和免疫对照组连续灌服生理盐水14 d,试验组连续灌服不同剂量AH(0.3、0.6和1.2 g/kg)14 d,分别在免疫后7、14、21、28 d采集血液、脾脏、胸腺和法氏囊等样品用于后续分析。结果显示:与空白对照组和免疫对照组相比,AH可以显著提高血清中新城疫血凝抑制(HI)抗体效价、特异性IgG抗体水平和肠道中特异性IgA抗体含量,提升血清中细胞因子白细胞介素2(IL-2)、IL-4、IL-17、干扰素γ(IFN-γ)水平;HE染色发现,AH可以显著促进脾脏、胸腺和法氏囊等免疫器官的发育;血生化分析发现AH对鸡肝肾功能无显著毒害作用。研究表明,AH可以作为一种安全的免疫增强剂,显著增强鸡的免疫功能和新城疫抗体水平。 In order to determine the effect of Alhagi honey polysaccharides(AH)on immune function and Newcastle disease(ND)antibody titer of chickens,1501-day-old Hylan brown roosters were obtained in this study and randomly divided into 5 groups with 30 rosters in each.When the maternal antibody was as low as log22.23,all chickens were immunized with the ND vaccine,except the Control group.The Control and ND groups were orally administered with normal saline for 14 days,and the experimental groups were orally administered with different doses of AH(0.3 g/kg,0.6 g/kg,1.2 g/kg,respectively)for 14 days.Samples such as blood,spleen,thymus and bursa of Fabricius were collected at 7,14,21 and 28 days after immunization,for subsequent analysis.The results showed that,compared with the Control and ND groups,AH remarkably increased the ND HI titer and the level of ND-specific IgG antibodies and cytokines(IL-2,IL-4,IL-17 and IFN-γ)in the serum and enhanced the level of ND-specific IgA in the intestines.HE staining showed that AH significantly improved the immune function of spleen,thymus and bursa of Fabricius.Blood biochemical analysis showed that AH had no significant toxic effect on the functions of the liver and kidney of the chickens.In conclusion,AH might be used as a safe immune-booster,which can significantly enhance the immune function and ND antibody titer of chickens.
