利用肿瘤基因图谱公共数据集分析膜外基质金属蛋白酶-14在肺癌中的表达及临床意义 被引量:1

Expression and clinical significance of matrix metalloproteinase-14 in lung cancer based on TCGA database

摘要 目的 通过肿瘤基因图谱(TCGA)数据库研究膜外基质金属蛋白酶-14(MMP14)在肺癌中的表达情况以及与肺癌临床病理特征的关系,进而推测MMP14对肺癌患者预后的影响,探讨MMP14在肺癌早期诊断中的作用。方法 检索TCGA中的肺癌数据集并下载资料,结合临床信息对芯片数据进行二次分析。采用R 3.6.3软件进行相关资料分析。结果 肺癌肿瘤组织中MMP14表达量高于正常组织(t=14.178,P<0.001)。在肺癌样本中,不同T分期的MMP14的表达量组间差异有统计学意义(P<0.001),T1、T2分期的MMP14表达量均比T3和T4低,早期肺癌的MMP14表达量与晚期相比有升高的趋势。Cox回归分析显示,TNM分期、MMP14和年龄是肺癌的独立危险因素。MMP14表达量与肺癌免疫细胞浸润的相关性分析显示,MMP14表达量与aDC、DC、iDC、单核细胞、中性粒细胞、NK细胞、TReg细胞、Th1和Th2细胞成正相关(P<0.001),与CD8 T细胞、嗜酸性粒细胞、Th17细胞成负相关(P<0.001),而与B细胞和T细胞无相关性(P>0.05)。结论 基于TCGA数据库分析发现MMP14在肺癌患者中高表达,在肺癌早期诊断中有一定的价值,并且与肺癌预后相关。MMP14在肺癌的早期诊断和预后中发挥着重要的作用,并可能成为预测肺癌预后的有用生物标志物。 Objective This paper aims to study the expression of extracellular matrix metalloproteinase-14(MMP14) in lung cancer and its relationship with clinicopathological features by using the cancer genome atlas(TCGA) database, so as to predict the effect of MMP14 on the prognosis of lung cancer patients and explore the role of MMP14 in the early diagnosis of lung cancer. Methods The lung cancer data set in TCGA was retrieved and downloaded, and the chip data were analyzed with clinical information. R 3.6.3 software was used to analyze relevant data. Results The expression of MMP14 in lung cancer tissues was higher than that in normal tissues(t=14.178, P<0.001). The levels of MMP14 in different T stages were different between groups, and the differences were statistically significant(P<0.001). The expression of MMP14 in T1 and T2 stages was lower than that in T3 and T4 stages. The expression of MMP14 in early stage was higher than that in advanced stage. Cox regression analysis showed that TNM stage, MMP14 and age were independent risk factors for lung cancer. Analysis of the correlation between MMP14 expression and immune cell infiltration in lung cancer showed that MMP14 expression was positively correlated with aDC, DC, iDC, monocytes, neutrophils, NK cells, TReg cells, Th1 and Th2 cells(P<0.001) and negatively correlated with CD8 T cells, eosinophils, Th17 cells(P<0.001), but not associated with B cells and T cells(P>0.05). Conclusion Based on TCGA database analysis, MMP14 is highly expressed in lung cancer patients, which has certain value in the early diagnosis of lung cancer, and it has correlation with the prognosis of lung cancer. MMP14 plays an important role in the early diagnosis and prognosis of lung cancer, and may be a useful biomarker for predicting the prognosis of lung cancer.
