宫颈癌预后相关长链非编码RNA筛选及其临床意义研究

摘要 目的 通过筛选宫颈癌预后相关长链非编码RNA(lncRNA),检测其在宫颈癌组织中的表达水平,并探讨其临床意义。方法 利用“edgeR”包筛选到与患者生存相关的差异表达lncRNA-AP001528.2。采用实时荧光定量聚合酶链反应技术检测AP001528.2的表达,并分析其与临床病理特征的相关性;生存曲线分析患者术后5年生存情况,Log-rank检测比较生存差异;Cox比例风险回归模型分析预后不良的独立危险因素;ROC曲线评价AP001528.2对宫颈癌的诊断价值。结果 宫颈癌组织中AP001528.2表达升高,且与FIGO分期、宫颈浸润深度和淋巴结转移相关;高表达AP001528.2的宫颈癌患者术后5年总生存率降低;FIGO分期、宫颈浸润深度、淋巴结转移和AP001528.2表达与宫颈癌患者生存率相关,且后三者是宫颈癌患者的独立危险因素。AP001528.2表达诊断宫颈癌ROC曲线下面积为0.84,敏感度为96.7%,特异度为60.7%。结论 AP001528.2可能是宫颈癌预后的独立不良因素之一,在宫颈癌的诊断中有一定的临床应用价值。 Objective This paper aims to screen long non-coding RNA(lncRNA) related to cervical cancer prognosis, detect its expression level in cervical cancer tissue, and analyze its clinical significance. Methods The differentially expressed lncRNA-AP001528.2 associated with patient survival was screened using the "edgeR" package. Real-time fluorescence quantitative polymerase chain reaction was used to detect the expression of AP001528.2, and its correlation with clinicopathological features was analyzed. The survival curve was used to analyze the patients’ survival rate at year 5 after surgery, and the survival difference was compared by Log-rank. Cox proportional hazard regression model was used to analyze the independent risk factors for poor prognosis. Diagnostic value of AP001528.2 in cervical cancer was evaluated by ROC curve. Results The expression of AP001528.2 was increased in cervical cancer tissue and correlated with FIGO stage, cervical invasion depth and lymph node metastasis. The 5-year overall survival rate of cervical cancer patients with high expression of AP001528.2 decreased after operation. FIGO stage, cervical invasion depth, lymph node metastasis and AP001528.2 expression are associated with the survival rate of cervical cancer patients, and the latter three are independent risk factors for cervical cancer patients. The area under the ROC curve of AP001528.2 in the diagnosis of cervical cancer was 0.84, the sensitivity was 96.7%, and the specificity was 60.7%. Conclusion AP001528.2 is one of the risk factors for poor prognosis of patients with cervical cancer, and it has certain clinical application value in the diagnosis of cervical cancer.
