氧化石墨烯对人类精子运动功能及形态影响的体外实验研究 被引量:2

In vitro experimental study about effects of graphene oxide on sperm motility and morphology

摘要 目的探讨氧化石墨烯(graphene oxide,GO)对体外精子运动功能及形态的影响。方法收集10例正常男性志愿者精子,上游法制备精子悬液。分别与不同浓度的GO(0、10、20和40μg/ml)共孵育1、3和6 h后,采用计算机辅助精子分析系统检测精子各项运动参数;采用Diff-Quik染色法分析精子形态。结果GO孵育1 h后,与对照组相比较,各剂量组精子总活力、前向运动百分率、精子直线速度、曲线速度及平均路径速度均无明显变化,差异无统计学意义(P>0.05)。孵育3 h后,与对照组相比较,高剂量组精子总活力及前向运动百分率明显降低,差异有统计学意义(P=0.016,P=0.022)。孵育6 h后,与对照组相比较,中和高剂量组精子总活力及前向运动百分率,高剂量组精子正常形态率均明显降低,差异均具有统计学意义(P<0.01),且精子总活力及前向运动百分率与GO剂量和作用时间呈负相关。结论GO对精子活力具有剂量和时间依赖性抑制作用,并影响精子正常形态,可能对精子运动功能产生不良影响。 Objective To investigate the effect of graphene oxide(GO)on sperm motility and morphology in vitro.Methods Sperm were collected from 10 normal male volunteers and prepared by upstream method.After incubation with different concentrations of GO(0,10,20,40μg/ml)for 1,3 and 6 h,The computer assisted sperm analysis system was used to detect the motility parameters of sperm,and sperm morphology was analyzed by Diff-Quik staining.Results Incubated for 1 h,no significant difference were found in the total motility,percentage of progressive motility,VSL,VCL and VAP between GO treated group and the control group(P>0.05).After incubation for 3 h,the total motility and percentage of progressive motility in the high-dose group were significantly decreased compared with the control group(P=0.016,P=0.022);After incubation for 6 h,the total motility and percentage of progressive motility in medium-dose and high-dose groups,as well as normal sperm morphology rate in high-dose group were significantly decreased compared with the control group(P<0.01).The total motility and percentage of progressive motility of sperm were negatively correlated with GO dose and exposure time.Conclusion GO has a dose-and time-dependent inhibitory effect on sperm motility,and affects the normal sperm morphology,which may have adverse effects on sperm motility.
