可摘局部義齒修復後口腔菌群的變化

摘要 目的臨床中可摘局部義齒(removable partial denture,RPD)戴入口腔後所產生多種口腔併發症均與原有的口腔微生態平衡狀態被打破有關,本文對戴入RPD後口腔菌群的變化進行研究,為口腔疾病的治療、預防提供參考。方法本文對10名成年患者和10名健康成年人在不同時間(治療前、治療後1月、治療後6月)的3種類型標本(唾液、齦上及齦下菌斑、黏膜菌群)進行了採集,使用變性梯度凝膠電泳(DGGE)對已擴增的目的片段進行檢測。結果重複數據方差分析(ANOVA)的統計結果顯示,戴入RPD後唾液和黏膜表面上菌群的種類數量在治療前後發生了改變。結論戴入RPD會使口腔菌群多樣性發生改變,口腔菌群有向着疾病的狀態發生轉變的趨勢。 Objective It is shown in clinical practice that wearing removable partial denture(RPD)has given rise to some oral diseases.The oral complications are commonly associated with the disruption of the equilibrium of oral microbial ecosystem which showed shift from healthy indigenous microflora community towards pathogenic microbial community.We studied the changes on the oral bacterial species before and after wearing RPD to provide guidance for the prevention and treatment of related oral diseases.Methods In this study,three kinds of samples(saliva,supra-and subgingival plaques,and oral mucosal surfaces)were collected from the 10-patients at three different times:before,1-month and 6-months after the treatment with RPD.Ten healthy adults were also selected as the control group.Denaturing gradient gel electrophoresis(DGGE)was applied to identify the bacterial profiles.Results The ANOVA of repeated measurement data indicated that,in saliva and mucosal surfaces,wearing RPDs significantly changed the numbers of amplicons,which showed the downward tendencies.Conclusion Wearing the RPDs influenced the diversity of bacterial species in the oral microbial ecosystem.It is noteworthy that the oral environment will be changed from the healthy status towards the disease status.
