基于荧光定量PCR的鹅绒定性检测方法

摘要 为了建立一种基于荧光定量PCR的鹅绒定性检测方法,以鹅线粒体基因CYTB基因序列设计引物和探针,进行荧光定量PCR扩增,以鸭绒、绵羊毛、兔毛、山羊绒、鸡毛、羊驼毛等纺织服装用蛋白质原材料作特异性检测,以提取的浓度为37.8 ng/μL的鹅绒DNA作为原液进行梯度稀释,进行了灵敏度检测、电泳重复性和扩增重复性验证。结果表明,该方法能够有效对鹅绒成分进行快速检测,具有较强的特异性,灵敏度较高,重复性良好。 To establish a method for detection of goose down based on PCR. The real-time PCR method for detection of goose down in down products was established using forward and reverse primers and probe corresponding to the mitochondrial gene. The specific detection was carried out by protein materials for textile and clothing including duck down, sheep wool, rabbit hair, cashmere, chicken feather, alpaca as reference animal species. Sensitivity detection was done using the 37.8 ng/μL goose down DNA as dope for gradient dilution. The electrophoretic repeatability and amplified repeatability were verified. This method can effectively detect goose down, with a strong specificity and high sensitivity.
