白藜芦醇激活Akt/eNOS信号缓解LPS诱导血管内皮细胞功能损伤的作用探讨 被引量:1

Research of Resveratrol Alleviates Vascular Endothelial Cell Injury Induced by LPS through Activating Akt/eNOS Signaling Pathway

摘要 探究白藜芦醇调控Akt/eNOS信号影响脂多糖(LPS)诱导的血管内皮细胞(VE)增殖、迁移和侵袭功能损伤。在VE细胞中添加不同浓度LPS(0,10,50,100,200,400,800和1 000μg/mL)和白藜芦醇(0,1,5,10,50,100,200和500μg/mL),确定LPS和白藜芦醇有效作用浓度,将VE细胞分为:Control组、LPS组(200μg/mL)和LPS+resveratrol组(200μg/mL LPS+50μg/mL白藜芦醇)。通过MTT法、划痕实验和Transwell小室实验评估白藜芦醇对LPS诱导的VE细胞增殖、迁移和侵袭能力的影响。Western blot检测VE细胞中p-Akt和p-eNOS表达。VE细胞活力随LPS和白藜芦醇处理浓度的增加而降低,在VE细胞中利用200μg/mL的LPS建立血管内皮细胞损伤模型,同时用50μg/mL的白藜芦醇处理。LPS组VE细胞的增殖(P<0.01)、迁移(P<0.01)和侵袭能力(P<0.01)均低于Control组。LPS+resveratrol组VE细胞的增殖(P<0.01)、迁移(P<0.01)和侵袭能力(P<0.01)均高于LPS组。同时,LPS组VE细胞的p-Akt(P<0.01)和p-eNOS(P<0.01)表达水平低于Control组,而LPS+resveratrol组VE细胞的p-Akt(P<0.01)和p-eNOS(P<0.01)表达水平高于LPS组。白藜芦醇可能通过增强Akt/eNOS信号活性,减轻LPS诱导的血管内皮细胞损伤,改善血管内皮功能。 To explore the regulation of resveratrol on vascular endothelial cell injury through activating Akt/eNOS signaling pathway, LPS(0,10,50,100,200,400,800 and 1 000 μg/mL) and resveratrol(0,1,5,10,50,100,200 and 500 μg/mL) were treated into VE cell.VE cells were divided into: Control group, LPS(200 μg/mL) group and LPS+resveratrol group(200 μg/mL LPS+50 μg/mL resveratrol).MTT,scratch and transwell assays were performed to examine the cell proliferation, migration and invasion of VE cell.Western blot was used to detect the expression levels of p-Akt and p-eNOS.The cell viability of VE cell were decreased with concentrations LPS and resveratrol.200 μg/mL LPS was applied into VE cell to establish a vascular endothelial cell injury model.50 μg/mL resveratrol was an optimal concentration.The VE cell proliferation(P<0.01),migration(P<0.01) and invasion(P<0.01) abilities of LPS group were lower than those of Control group.While the VE cell proliferation(P<0.01),migration(P<0.01) and invasion(P<0.01) abilities of LPS+resveratrol group were higher than those of LPS group.Meanwhile, the expression levels of p-Akt(P<0.01) and p-eNOS(P<0.01) in VE cell were inhibited in LPS group as compared to Control group.The expression levels of p-Akt(P<0.01) and p-eNOS(P<0.01) in VE cell were increased in LPS+resveratrol group as compared to LPS group.Resveratrol alleviates vascular endothelial cell injury by activating Akt/eNOS signaling pathway, and enhances vascular endothelial function.
