LncRNA TUG1靶向miR-141-3p影响甲状腺癌细胞的增殖和侵袭

Effect of LncRNA TUG1 on proliferation and invasion of thyroid carcinoma cells by targeting miR-141-3p

摘要 目的检测LncRNA TUG1在甲状腺癌细胞中的表达,初步探讨LncRNA TUG1对甲状腺癌细胞增殖和侵袭的影响及机制。方法采用qRT-PCR法检测甲状腺癌B-CPAP、TPC-1细胞株中LncRNA TUG1和miR-141-3p的表达水平,应用生物信息学预测lncRNA TUG1和miR-141-3p的靶向关系,双荧光素酶报告基因实验进行验证。将lncRNA TUG1干扰序列转染TPC-1细胞,应用qRT-PCR法检测lncRNA TUG1和miR-141-3p的表达,MTT法检测各组TPC-1细胞增殖能力,Transwell实验检测各组TPC-1细胞的侵袭能力。结果与正常甲状腺上皮细胞相比,BCPAP、TPC-1中LncRNA TUG1表达水平明显升高(P<0.05),miR-141-3p表达水平明显下降(P<0.05),生物信息学分析及双荧光素酶实验结果显示,lncRNA TUG1可以靶向调控miR-141-3p。转染TUG1干扰序列能够降低TPC-1细胞lncRNA TUG1的表达,上调miR-141-3p的表达(P<0.05);沉默lncRNA TUG1表达,TPC-1细胞的增殖与侵袭能力下降(P<0.05)。结论沉默lncRNA TUG1可以抑制甲状腺癌细胞TPC-1的增殖和侵袭,其机制可能与上调miR-141-3p表达有关。 Objective To detect the expression of lncRNA TUG1 in thyroid carcinoma cells,and explore its effect on cell proliferation and migration.Methods The levels of lncRNA TUG1 and miR-141-3p in thyroid carcinoma B-CPAP and TPC-1 cell lines were detected by qRT-PCR.Bioinformatics was used to predict the targeting relationship between lncRNA TUG1 and miR-141-3p,which was verified by the dual luciferase reporter gene experiment.The TPC-1 cells were cultured in vitro and transfected with the Si-TUG1.The levels of lncRNA TUG1 and miR-141-3p were detected by quantitative real-time PCR.The proliferation ability of TPC-1 cells was detected by MTT.The invasion ability of TPC-1 cells was detected by Transwell.Results The level of lncRNA TUG1 in BCPAP and TPC-1 cells was significantly higher than that in normal thyroid epithelial cells(P<0.05).Bioinformatics and double-luciferase assay confirmed that TUG1 can regulate the expression of miR-141-3p.After silencing lncRNA TUG1,the expression of lncRNA TUG1 and was decreased and the expression of miR-141-3p was up-regulated.The inhibition of lncRNA TUG1 reduced the proliferation and invasion of TPC-1 cells.Conclusion Silencing lncRNA TUG1 can inhibit the proliferation and invasion of TPC-1 cells,and its mechanism may be related to the targeted regulation of miR-141-3p expression.
