摘要
目的探究大蒜素对过敏性气道炎症的治疗作用及对cAMP/PKA/CREB通路的影响。方法40只SPF级雄性BALB/c小鼠,随机分为空白组、模型组、大蒜素组、地塞米松组,每组10只。模型组与给药组利用卵白蛋白(OVA)致敏联合雾化激发的方法建立过敏性哮喘模型。检测炎症细胞数量变化;HE染色、Masson染色观察肺组织病理改变及气道重塑情况;ELISA检测支气管肺泡灌洗液(BALF)中IL-4、IL-5、IL-13、IFN-γ、TNF-α及IgE水平变化;免疫荧光方法观察肺组织cAMP蛋白表达变化;Western blot检测肺组织PKA、CREB、p-CREB蛋白表达变化。结果大蒜素能够改善肺组织炎症病理损伤,调节气道重塑;降低BALF中炎性细胞数量、降低IL-4、IL-5、IL-13、TNF-α表达、降低IgE含量,上调IFN-γ表达;上调肺组织cAMP、PKA、CREB、p-CREB蛋白的表达。结论大蒜素可能通过调控cAMP/PKA/CREB信号通路抑制过敏性哮喘小鼠气道炎症反应。
Objective To explore the therapeutic effect of allicin on allergic airway inflammation and its effect on cAMP/PKA/CREB pathway.Methods Forty SPF male BALB/c mice were randomly divided into blank group,model group,allicin group and dexamethasone group,with 10 mice in each group.The allergic asthma model was established by ovalbumin(OVA)sensitization combined with atomization stimulation in the model group and administration group.The number of inflammatory cells was detected.The pathological changes of lung tissue and airway remodeling were observed by HE staining and Masson staining.The levels of IL-4,IL-5,IL-13,IFN-γ,TNF-αand IgE in bronchoalveolar lavage fluid(BALF)were determined by ELISA.The expression of cAMP in lung tissues was observed by immunofluorescence method.The expressions of PKA,CREB and p-CREB in lung tissues were detected by Western blot.Results Allicin can improve lung tissue inflammation and pathological injury and regulate airway remodeling.The number of inflammatory cells in BALF was decreased.The expression of IL-4,IL-5,IL-13,TNF-αwas decreased.The IgE content was decreased.The expression of IFN-γwas up-regulated.The expressions of cAMP,PKA,CREB and p-CREB in lung tissue was up-regulated.Conclusion Allicin may inhibit airway inflammation in mice with allergic asthma by regulating cAMP/PKA/CREB signaling pathway.
