小檗碱调控HIF-1α和TGF-β抑制鼻咽癌细胞增殖和侵袭转移

Berberine inhibits nasopharyngeal carcinoma cancer cell proliferation,invasion and metastasis by regulating the expression of HIF-1α and TGF-β

目的探讨小檗碱对人鼻咽癌细胞5-8F细胞增殖和侵袭转移的影响及可能机制。方法体外培养人鼻咽癌细胞5-8F细胞后,将细胞分为对照组(0μmol/L)、12.5μmol/L、25μmol/L和50μmol/L小檗碱处理组。CCK-8检测5-8F细胞的增殖情况;划痕实验检测鼻咽癌细胞迁移能力;穿梭小室实验检测鼻咽癌细胞侵袭能力的变化;Western blot检测各组细胞中HIF-1α信号通路相关蛋白和TGF-β的表达。结果小檗碱抑制5-8F细胞增殖,并呈浓度依赖性和时间依赖性。划痕实验和穿梭小室实验结果表明小檗碱能显著降低5-8F细胞迁移和侵袭能力。Western blot检测结果发现,HIF-1α和TGF-β的表达在药物作用后显著降低。结论小檗碱通过抑制HIF-1α和TGF-β的表达抑制人鼻咽癌细胞5-8F细胞增殖、迁移和侵袭。

Objective To investigate the effect and possible mechanism of berberine on the proliferation,invasion and metastasis of human nasopharyngeal carcinoma 5-8F cells.Methods After cultured in vitro,human nasopharyngeal carcinoma 5-8F cells were divided into control groups(treated with 0μmol/L),12.5μmol/L,25μmol/L and 50μmol/L berberine group.CCK-8 assay was performed to detect cell proliferation.The scratch examination was used to examine the capability of cell migration.Transwell assay was performed to detect capability of cell invasion.Western blot was used to detect the expression of HIF-1α and TGF-β.Results Berberine could inhibit the proliferation of 5-8F cells in a concentration-dependent and time-dependent manner.The results of scratch test and shuttle chamber test showed that berberine could significantly reduce the migration and invasion ability of 5-8F cells.Western blot analysis showed that the expressions of HIF-1α and TGF-β were significantly decreased after drug treatment.Conclusion Berberine inhibits the proliferation,migration and invasion of human nasopharyngeal carcinoma 5-8F cells by inhibiting the expression of HIF-1α and TGF-β.
