摘要
目的探究泛素样含PHD和环指域蛋白1(UHRF1)在低级别胶质瘤中表达水平及与患者预后之间的联系。方法从TCGA数据库中下载提取低级别胶质瘤样本和在GTEx数据库中的正常脑组织样本的转录组数据合并后分析UHRF1的表达水平;分析比较UHRF1表达水平高低不同的低级别胶质瘤患者组的预后区别;GSEA富集分析UHRF1在低级别胶质瘤中主要参与的信号通路;通过TIMER数据库分析低级别胶质瘤组织中UHRF1表达与常见的免疫细胞浸润的相关性;选取6例低级别胶质瘤组织及3例非癌脑组织,通过RT-qPCR的方法验证UHRF1在低级别胶质瘤与非癌脑组织的表达情况。结果UHRF1在低级别胶质瘤组织中的表达量明显高于非癌脑组织,且UHRF1表达水平高的患者在预后相对更差(P<0.01);富集分析显示,UHRF1可能通过MYC_TARGETS_V2通路在低级别胶质瘤中发挥作用;在低级别胶质瘤组织中,UHRF1的表达与巨噬细胞呈正相关(P<0.01);在收集到的病例中,与非癌脑组织相比,UHRF1在低级别胶质瘤中的表达显著增高(P<0.001)。结论UHRF1在胶质瘤中高表达且有着更差的预后,其可能成为治疗低级别胶质瘤的潜在靶点。
Objective To investigate the expression levels of ubiquitin-like PHD-containing and ring finger domain protein 1(UHRF1)in low-grade gliomas and the association with patient prognosis.Methods The transcriptomic data of low-grade glioma samples from TCGA database and normal brain tissue samples from GTEx database were combined to analyze the level of UHRF1.The prognosis of low-grade glioma patients with different levels of UHRF1 was compared.The main signaling pathways involved in UHRF1 in low-grade glioma were analyzed by GSEA enrichment.The level of UHRF1 in low-grade glioma was analyzed by TIMER database.TIMER database to analyze the correlation between UHRF1expression in low-grade glioma tissues and common immune cell infiltration.Six cases of low-grade glioma tissues and three cases of non-cancerous brain tissues were selected to verify the expression of UHRF1 in low-grade glioma and noncancerous brain tissues by RT-qPCR.Results The expression of UHRF1 in low-grade glioma tissues was significantly higher than that in non-cancerous brain tissues,and patients with high UHRF1 expression levels were relatively worse in prognosis(P<0.01).Enrichment analysis showed that UHRF1 may play a role in low-grade glioma through the MYC_TARGETS_V2 pathway.The expression of UHRF1 in low-grade glioma tissues was positively correlated with macrophage.The expression of UHRF1 was positively correlated with macrophages in low-grade glioma tissues(P<0.01).In the collected cases.the expression of UHRF1 was significantly higher in low-grade gliomas compared with non-cancerous brain tissues(P<0.001).Conclusion UHRF1 is highly expressed in glioma and has a worse prognosis,which may be a potential target for the treatment of low-grade glioma.
