超声波处理对小麦种子活力影响的转录组分析 被引量:4

摘要 为分析超声波处理对小麦种子活力及相关基因表达的影响,设置不同时长超声波(0(CK)、10、20、30、40和50min)处理小麦种子,同时借助转录组测序技术筛选与小麦种子活力相关的差异表达基因,进行GO分类及KEGG富集分析。结果表明,超声波处理30min的小麦种子活力最高,且显著高于其他处理和CK。30min处理的小麦幼苗萌发0h的脯氨酸含量以及SOD、CAT、POD的活性均显著高于CK,萌发72h的幼苗SOD、CAT活性与脯氨酸含量均显著高于CK。转录组分析表明,与CK相比,超声波处理30min的小麦种子萌发0和72h共有1723个差异表达的基因。GO富集分析表明,在生物过程上,超声波处理对种子活力的影响主要集中在氧化还原过程、碳水化合物代谢过程、氧化应激响应与过氧化氢分解代谢过程;在细胞组分上,主要集中在胞外区、质外体、细胞壁和细胞外间隙等质膜外区域;在分子功能上,主要集中在氧化还原酶活性、单加氧酶活性和过氧化物酶活性。KEGG富集分析发现,富集最为显著的3个通路依次为谷胱甘肽代谢(ko00480)、代谢途径(ko01100)与色氨酸代谢(ko00380),其中谷胱甘肽代谢通路共富集32个差异表达基因(DEGs)。 In order to analyze the effect of ultrasonic treatment duration on wheat seed vigor and related gene expression,difference ultrasonic treatment times(0(CK),10,20,30,40and 50min)were designed to pre-treat wheat seeds.Based on RNA-seq analysis,this study chose the differentially expressed genes that enhanced seed vigor by ultrasonic,and used these genes for GO classification and KEGG enrichment analysis.The result showed that the wheat seed vitality of 30min treatment was the highest,and have significant difference with other treatments.The30min ultrasonic treatment significantly increased the proline content,SOD,CAT and POD activities of wheat seedlings than those of CK group at the germination time point of 0h.Meanwhile,the SOD,CAT and proline contents of seedlings 72hafter germination were significantly higher than those of untreated control.Transcriptome analysis showed that there were 1723DEGs between 0min control group and 30min ultrasonic treatment group at germination of 0and 72h.GO enrichment analysis showed that the regulation of seed vigor under ultrasonic treatment mainly concentrated on oxidation-reduction process,carbohydrate metabolic process,response to oxidative stress and hydrogen peroxide catabolic process in terms of biological process,and extracellular region,apoplast,cell wall and extracellular space in terms of cell components,meanwhile,oxidoreductase activity,monooxygenase activity and peroxidase activity in terms of molecular function.KEGG enrichment analysis showed that the three pathways with the most significant enrichment were glutathione metabolism(ko00480),metabolic pathway(ko01100)and tryptophan metabolism(ko00380),respectively.Among them,a total of 32differentially expressed genes(DEGs)were enriched in glutathione metabolism pathway.
