高压氧联合骨髓间充质干细胞移植对脊髓损伤大鼠PERK表达、炎性因子及氧化应激指标的影响 被引量:1

Effects of hyperbaric oxygen combined with bone marrow mesenchymal stem cell transplantation on PERK expression,inflammatory factors,and stress indicators in rats with spinal cord injury

摘要 目的:探究高压氧(HBO)联合骨髓间充质干细胞(BMSCs)移植对脊髓损伤(SCI)大鼠蛋白激酶R样内质网激酶(PERK)表达、炎性因子及氧化应激相关指标的影响。方法:选取30只Sprague Dawley雄性大鼠为研究对象,均建立SCI模型,依据随机数字表法分为模型组、BMSCs组、HBO联合BMSCs组(BMSCs+HBO组),每组10只。观察各组大鼠PERK表达、炎性因子及各时间点(SCI损伤后12 h及1、3、7、14 d)氧化应激指标的变化情况。结果:与模型组比较,BMSCs组、BMSCs+HBO组大鼠的PERK蛋白水平均降低,且BMSCs+HBO组降幅显著大于BMSCs组,差异有统计学意义(P<0.05)。与模型组比较,BMSCs组、BMSCs+HBO组大鼠的血清TNF-α、IL-6、IL-10水平均降低,BMSCs+HBO组降幅明显大于BMSCs组(P<0.05)。SCI损伤后7 d,BMSCs+HBO组大鼠的血清MDA水平明显低于BMSCs组(P<0.05)。BMSCs+HBO组大鼠损伤后3、7、14 d的血清SOD水平明显高于BMSCs组(P<0.05)。结论:SCI大鼠的PERK表达活跃,炎性因子水平增高,HBO联合BMSCs可有效抑制PERK蛋白表达,降低炎性因子水平,改善氧化应激指标,对脊髓修复具有促进作用。 Objective:To explore the effects of hyperbaric oxygen combined with bone marrow mesenchymal stem cells(BMSCs)transplantation on the expression of protein kinase R-like endoplasmic reticulum kinase(PERK),inflammatory factors,and oxidative stress-related indicators in rats with spinal cord injury(SCI).Methods:A total of thirty Sprague Dawley male rats were selected as the research objects.SCI models were established and divided into model group,BMSCs group,and HBO combined with BMSCs group(BMSCs+HBO group)according to the random number table method,with 10 rats in each group.The changes of PERK expression,inflammatory factors,and stress indicators at each time points(at 12 h after SCI injury,and 1,3,7,and 14 d after SCI injury)in the three groups were observed.Results:Compared with the model group,the PERK protein levels of the rats in the BMSCs group and the BMSCs+HBO group were decreased,and the decrease in the BMSCs+HBO group was significantly greater than that in the BMSCs group(P<0.05).Compared with the model group,the serum levels of TNF-α,IL-6,and IL-10 in the BMSCs group and the BMSCs+HBO group were decreased,and the decrease in the BMSCs+HBO group was significantly greater than that in the BMSCs group,and the difference was statistically significant(P<0.05).On day 7 after SCI injury,the serum MDA level in the BMSCs+HBO group was significantly lower than that in the BMSCs group,and the difference was statistically significant(P<0.05).The serum SOD level in the BMSCs+HBO group was significantly higher than those in the BMSCs group on day 3,7,and 14 after injury,and the differences were statistically significant(P<0.05).Conclusion:The expression of PERK is active in SCI rats,and the level of inflammatory factors is increased.HBO combined with BMSCs can effectively inhibit the expression of PERK,reduce the level of inflammatory factors,improve oxidative stress indicators,thus promoting spinal cord repair.
