基于PI3K/Akt/Nrf2信号通路探讨小蓟饮子治疗小鼠放射性膀胱炎作用机制 被引量:1

Mechanism of Xiaoji Yinzi in Treating Acute Radiation Cystitis in Mice Based on PI3K/Akt/Nrf2 Signal Pathway

摘要 目的基于PI3K/Akt/Nrf2信号通路探讨小蓟饮子治疗小鼠放射性膀胱炎(RC)的作用机制。方法将30只健康雌性SPF级ICR小鼠采用随机数字表法分为11只空白组和19只造模组。空白组不予造模,造模组腹腔麻醉后暴露膀胱部位于X射线辐照仪下以6-MV X射线、2 Gy/min照射10 min建立RC小鼠模型,空白组和造模组各随机取3只处死后,在光镜下观察膀胱组织形态学改变以确定造模成功。造模成功后将造模组按随机数字表法分为模型组和中药组各8只,中药组按10 mL/(kg·d)灌胃1.68 g/mL小蓟饮子浓缩液,空白组和模型组灌胃等体积生理盐水,连续灌胃7 d。灌胃结束后使用酶联免疫吸附测定(ELISA)法测定小鼠眼球血氧化应激指标[超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC);实时荧光定量PCR(RT-PCR)法检测膀胱组织中磷脂酰肌醇-3-激酶(PI3K)、蛋白激酶B(Akt)、核因子E2相关因子2(Nrf2)、血红素氧合酶-1(HO-1)mRNA相对表达水平。结果与空白组比较,模型组小鼠眼球血中SOD、T-AOC、GSH-Px含量均降低(P均<0.05),MDA含量升高(P<0.05),膀胱组织中PI3K、Akt、Nrf2、HO-1 mRNA相对表达水平均升高(P均<0.05);与模型组比较,中药组小鼠眼球血中SOD、T-AOC、GSH-Px含量均升高(P<0.01或P<0.05),MDA含量降低(P<0.05),膀胱组织中PI3K、Akt、Nrf2、HO-1 mRNA相对表达水平均升高(P均<0.01)。结论小蓟饮子可有效改善RC小鼠模型的损伤,其作用机制可能与PI3K/Akt/Nrf2信号通路调控氧化应激有关。
