摘要
目的探索转录延伸因子A样3因子(TCEAL3)对食管鳞癌TE-1和KYSE-30细胞系增殖、迁移、凋亡的影响。方法将食管鳞癌细胞TE-1和KYSE-30细胞株分成TE-1对照组和KYSE-30对照组(均转染空载慢病毒)、TE-1实验组和KYSE-30实验组(均转染TCEAL3过表达慢病毒)。以蛋白质印迹法检测TCEAL3、高迁移率族蛋白A1(HMGA1)和基质金属蛋白酶2(MMP2)的表达水平;以CCK8实验检测细胞的增殖情况;以Transwell迁移实验检测细胞迁移情况;以流式细胞术检测细胞凋亡情况。结果TE-1对照组和TE-1实验组、KYSE-30对照组和KYSE-30实验组的TCEAL3蛋白相对表达水平分别为0.31±0.02和0.56±0.02,0.25±0.02和0.49±0.05;细胞增殖率分别为(1721.01±21.16)%和(1229.69±59.17)%,(1715.27±89.70)%和(781.05±136.75)%;凋亡率分别为(3.99±1.60)%和(32.20±2.96)%,(5.15±0.20)%和(32.20±0.53)%;迁移细胞数分别为(189.00±6.67)和(120.00±28.40)个,(189.00±30.20)和(111.00±27.60)个;HMGA1蛋白表达水平分别为0.79±0.11和0.37±0.06,0.67±0.03和0.33±0.03;MMP2蛋白表达水平分别为0.55±0.02和0.32±0.04,0.61±0.02和0.37±0.01。上述指标,TE-1实验组与TE-1对照组比较,KYSE-30实验组与KYSE-30对照组比较,差异均有统计学意义(均P<0.05)。结论TCEAL3可能通过抑制HMGA1的表达,从而下调MMP2的表达来抑制食管鳞癌细胞的增殖能力和迁移运动能力,并促进凋亡。
ObjectiveTo investigate the effects of Transcription elogation factor A like 3 (TCEAL3) on proliferation,migration,apoptosis of esophageal squamous cell carcinoma TE-1 and KYSE-30cell lines.Methods The esophageal squamous cell TE-1 and KYSE-30 cell lines were divided into TE-1 control group and KYSE-30 control group (no-loaded lentiviral transfection) and TE-1experimental group and KYSE-30 experimental group (TCEAL3overexpression lentiviral transfection).The protein expression of TCEAL3,high mobility group protein A1 (HMGA1) and matrix metallopeptidase 2 (MMP2) were detected by Western blotting;Cell counting kit-8 was used to detect cell proliferation;the Transwell method was used to detect cell invasion ability;flow cytometry was used to detect cell apoptosis.Results The relative expression of TCEAL3 protein in the TE-1 control group,TE-1experimental group and KYSE-30 control group,KYSE-30 experimental group were 0.31±0.02 and 0.56±0.02,0.25±0.02 and 0.49±0.05,respectively;the cell proliferation rates were (1 721.01±21.16)%and(1 229.69±59.17)%,(1 715.27±89.70)%and (781.05±136.75)%,respectively;the apoptosis rates were(3.99±1.60)%and (32.20±2.96)%,(5.15±0.20)%and (32.20±0.53)%,respectively;the number of migrated cells was 189.00±6.67 and 120.00±28.40,189.00±30.20 and 111.00±27.60,respectively;HMGA1protein expression is as follows 0.79±0.11 and 0.37±0.06,0.67±0.03 and 0.33±0.03,respectively;the amount of MMP2 protein expression is as follows 0.55±0.02 and 0.32±0.04,0.61±0.02 and 0.37±0.01,respectively.There were statistically significant differences in the above indexes between TE-1 control group and TE-1experimental group,between KYSE-30 control group and KYSE-30 experimental group (all P<0.05).Conclusion TCEAL3 may inhibit the proliferation and migration of esophageal squamous cell carcinoma cells by inhibiting the expression of HMGA1 and thereby downregulating the expression of MMP2,and promote apoptosis.
