摘要
为研究4种蔷薇属植物(毛萼蔷薇、大花香水月季、维西蔷薇、丽江蔷薇)的抗寒性差异,以筛选出耐寒性强的品种,利用低温培养箱进行不同梯度低温(0、-5、-10、-15℃)胁迫处理,测定4种蔷薇属植物可溶性糖含量、可溶性蛋白含量、丙二醛含量、脯氨酸(Pro)含量、SOD活性、POD活性及耐寒相关调控基因HSP90的表达差异。结果表明,在低温胁迫下,4种蔷薇属植物叶片可溶性蛋白含量、可溶性糖含量、游离脯氨酸含量、丙二醛含量、SOD活性和POD活性均随低温胁迫加剧呈现逐渐升高趋势。对4种蔷薇属植物HSP90基因的差异表达分析表明,该基因在4种植物中的相对表达量随低温胁迫加剧呈现逐渐升高趋势,其中,毛萼蔷薇在0、-5、-10、-15℃低温胁迫下HSP90基因表达量均最高。表明4种蔷薇属植物的抗寒性存在差异,其中,毛萼蔷薇的抗寒性较好。
To study the difference of cold resistance of four plants of Rosa(R. lasiosepala, R. odorata. var. gigantea, R.weisiensis, R. lichiangensis), and screen out the varieties with strong cold resistance. In this study, a low temperature incubator was used to conduct different gradients of low temperature stress(0,-5,-10,-15 ℃), the soluble sugar content, soluble protein content, malondialdehyde(MDA) content, proline(Pro) content, SOD activity, POD activity, and expression difference of coldresistance-related regulatory gene HSP90 were measured. The results showed that under low temperature stress, the soluble protein content, soluble sugar content, free proline content, malondialdehyde content, SOD activity, and POD activity in the leaves of four plants of Rosa showed a gradually increasing trend with aggravation of low temperature stress. The analysis of the differential expression of HSP90 gene of four plants of Rosa showed that the relative expression of HSP90 gene showed a trend of gradual increase with aggravation of low temperature stress. The expression of HSP90 gene was the highest in R. lasiosepala under 0,-5,-10,-15 ℃ low temperature stress, indicating that there were differences in cold resistance of four plants of Rosa,among which R. lasiosepala had better cold resistance.
