摘要
目的:探讨高剂量维生素C(VC)对肝细胞癌的抑制作用及其作用机制。方法:培养人肝癌细胞系Huh7细胞,取对数增殖期细胞用于实验。对照组加入不含VC的培养基;实验组加入不同浓度的VC处理,CCK8法筛选半数抑制剂量4 mM。分别检测对照组和4 mM VC组(以下称为VC组)的Huh7细胞增殖、细胞周期和凋亡等情况。结果:VC干预后,显著降低Huh7细胞的增殖活性,VC组的细胞增值能力显著降低,差异具有统计学意义(P<0.01);Tunel检测VC组的细胞大量凋亡;流式细胞术检测VC组的细胞凋亡增加,细胞周期G1期变长,差异具有统计学意义(P<0.01);TRANSWELL法检测VC组细胞迁移和侵袭能力下降,差异具有统计学意义(P<0.01)。结论:4 mM浓度的VC对肝癌细胞Huh7具有抑制作用,其作用机制与增加细胞凋亡和增长细胞周期作用有关。
Objective:To investigate the inhibitory effect of high-dose vitamin C(VC)on hepatocellular carcinoma and its mechanism.Methods:Human hepatoma cell line Huh7 cells were cultured and cells in logarithmic proliferative phase were taken for experiments.The control group was added with medium without VC;the experimental group was treated with different concentrations of VC,and CCK8 method was used to screen the half inhibitor dose of 4 mM.The proliferation,cell cycle and apoptosis of Huh7 cells in control group and 4 mM VC group(hereinafter referred to as VC group)were detected.Results:After VC intervention,the proliferation activity of Huh7 cells was significantly decreased,and the proliferation ability of VC group was significantly decreased(P<0.01).The apoptosis of VC group was detected by Tunel.The apoptosis and G1 phase of cell cycle were detected by flow cytometry(P<0.01).The migration and invasion ability of VC group were significantly decreased by TRANSWELL method(P<0.01).Conclusion:VC at a concentration of 4mM had an inhibitory effect on hepatoma cells Huh7,whose mechanism of action is related to increasing apoptosis and cell cycle.
