芒柄花苷对小鼠脓毒症引起认知功能障碍的影响

摘要 目的:探讨芒柄花苷对小鼠脓毒症引起认知功能障碍的影响。方法:选取60只8周龄雄性C57BL/6小鼠,采用随机数字表法分为假手术组(C组)、脓毒症组(CLP组)、芒柄花苷(ON组)和芒柄花苷+脓毒症组(ON+CLP组),每组各15只。采用盲肠结扎穿孔(CLP)制备脓毒症模型。ON组与ON+CLP组在造模前进行芒柄花苷(30 mg/kg)灌胃,1次/d,连续7 d;C组与CLP组每日给予等剂量生理盐水。造模后比较各组小鼠7 d生存率;Morris水迷宫实验检测各组小鼠认知功能,比较各组小鼠海马组织中肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)、IL-6、超氧化物歧化酶(SOD)、丙二醛(MDA)水平。结果:与C组比较,CLP组小鼠生存率降低(P<0.05);与CLP组比较,ON+CLP组小鼠生存率增加(P<0.05)。与C组比较,CLP组小鼠逃避潜伏期延长,穿越平台次数减少,待在平台所在象限时间缩短(均P<0.05);与CLP组比较,ON+CLP组小鼠逃避潜伏期缩短,穿越平台次数增多,待在平台所在象限时间延长(均P<0.05)。与C组比较,CLP组小鼠TNF-α、IL-1β、IL-6及MDA含量升高,SOD活性降低(均P<0.05);与CLP组比较,ON+CLP组TNF-α、IL-1β、IL-6和MDA含量降低,SOD活性增加(均P<0.05)。结论:芒柄花苷可以改善小鼠脓毒症引起的认知功能障碍,机制可能与芒柄花苷抑制海马组织的炎症反应和氧化应激反应有关。 Objective:To investigate the effect of ononin on the cognitive function of sepsis mice.Methods:60 male C57BL/6 mice aged 8 weeks were divided into four groups(15 in each group):control group(C Group),sepsis group(CLP group),ononin group(ON group),and ononin+CLP group(ON+CLP group) by digital random table methods, 15 cases in each group.Cecal ligation and perforation(CLP) was used to establish the sepsis model.The ON group and the ON+CLP group were given ononin(30 mg/kg) once a day for 7 days before modeling, group C and CLP were given equal dose of normal saline every day.The 7-day survival rate of mice was observed after modeling.Morris water maze(MWM) test was employed to detect the cognitive function of mice in each group.The levels of TNF-α,IL-1β,IL-6,SOD and MDA in hippocampus tissues of mice were compared between the groups.Results:Compared with C group, the survival rate of CLP group was lower(P<0.05).The survival rate of ON+CLP group was increased relative to CLP group(P<0.05).The escape latency was prolonged, the times of crossing the platform were reduced, and the time to staying in the quadrant of the platform was shortened in CLP group when compared with C group(all P<0.05).Compared with CLP group, the escape latency was significantly shortened, the number of times of crossing the platform was enhanced, and the time of staying in the quadrant of the platform was prolonged for ON+CLP group(all P<0.05).Compared with C group, the content of TNF-α,IL-1β,IL-6 and MDA in hippocampus of CLP group was remarkably increased, while the activity of SOD exhibited decreased(all P<0.05).Compared with CLP group, the content of TNF-α,IL-1β,IL-6 and MDA in ON+CLP group decreased, and the activity of SOD exhibited increased(all P<0.05).Conclusion:Ononin prophylaxis could ameliorate the cognitive decline of sepsis mice, and the underlying mechanism may be related to its anti-inflammatory and antioxidant effects.
