多肉植物潘灯玉露快繁体系研究

摘要 为明确百合科十二卷属潘灯玉露在植物组织培养中的最适激素配比,建立多肉快繁技术体系。本研究将玉露叶片作为离体培养材料,设置了含不同浓度植物生长调节剂的MS和1/2MS培养基,研究了NAA、6-BA和KT对外植体脱分化和再分化等多环节的影响。实验结果显示,当pH=5.8时,MS+0.1 mg/mL NAA+0.5 mg/mL 6-BA的脱分化诱导效果最好,对胚性愈伤组织的诱导率达66.7%,并可形成净增殖量为0.761 g的晶体状胚性愈伤组织;经初代培养的愈伤组织在MS+0.1 mg/mL NAA+0.5 mg/mL 6-BA+0.5 mg/mL KT上56 d后,可以诱导产生净增殖量为12.115 g的愈伤组织;当愈伤组织转接至MS+0.5 mg/mL NAA中可产生高达93.3%的丛芽分化率;此外,幼苗在1/2 MS+0.5 mg/mL NAA上生根率也超过了85%。综上,在潘灯玉露实际生产应用中,建议使用以上浓度配方对外植体进行愈伤组织诱导、扩增、丛芽的分化和生根处理。 In order to determine the optimal hormone ratio of liliaceae in plant tissue culture,the rapid propagation technology system of succulent Haworthia was established.In this study,the leaves of Haworthia were used as in vitro culture materials,and MS and 1/2MS medium containing different concentrations of plant growth regulators were set up.The effects of NAA,6-BA and KT on dedifferentiation and redifferentiation of explants were studied.The results showed that when pH=5.8,MS+0.1mg/ml NAA+0.5mg/ml 6-BA had the best effect on cell dedifferentiation induction,the induction rate of embryonic callus reached 66.7%,and the net proliferation amount of crystalline embryonic callus was 0.761 g.After 56 d of MS+0.1 mg/ml NAA+0.5 mg/ml 6-BA+0.5 mg/ml KT,induced callus with a net proliferation of 12.115 g.When callus were transferred to MS+0.5 mg/ml NAA,the differentiation rate of plexus bud was 93.3%.The seedlings could produce more than 85%rooting rate on 1/2 MS+0.5 mg/ml NAA.In conclusion,in the practical production and application of Haworthia cooperivar,it is recommended to use the above concentration formula for callus induction,callus amplification,bush bud differentiation and rooting treatment of explants.
