黄芩苷对ARPE-19细胞氧化应激损伤的保护作用 被引量:3

摘要 目的本研究旨在揭示黄芩苷对氧化应激状态下视网膜色素上皮细胞的保护作用。方法AMD模型构建及氧化应激损伤评估;实验组、对照组及黄芩苷干预组分别作用于细胞,检测细胞衰老状态,ROS(reactive oxidative species)累积,细胞增殖、凋亡(apoptosis)状态以及Real-Time定量荧光聚合酶链反应分析SIRT1 mRNA水平。结果1.氧化应激能造成ARPE损伤,并导致SIRT1 mRNA表达水平下降,差异有统计学意义(P<0.05)。2.黄芩苷高、低剂量组能显著改善细胞衰老状态,促进增殖,降低凋亡,并显著改善SIRT1 mRNA水平,差异有统计学意义(P<0.05)。结论ARPE细胞氧化应激损伤中黄芩苷能起到负向调节作用,意味着黄芩苷具有抗氧化应激损伤的作用,为临床AMD治疗提供了思路。
