蚊母树叶瘿蚜分子鉴定及种群遗传分化研究

摘要 为了明确我国蚊母树叶片虫瘿致瘿蚜虫的种类及其种群遗传分化情况,从湖南、重庆、江苏、浙江4地区的蚊母树叶瘿蚜标本中提取DNA进行线粒体COI、COII基因片段序列分析,结合GenBank同源序列进行了分子鉴定,并构建了基于COI、COII基因序列的蚜虫分子系统进化树。结果表明:南京中山植物园蚊母树叶瘿蚜基因片段序列与湖南省长沙、娄底、永州种群的完全相同,即共享1个单倍型;重庆、杭州地理种群存在一定的遗传分化,但仍以长沙、南京标本的单倍型为主,且各单倍型样本之间的遗传距离非常小。序列分析结果支持4地区6个城市的蚊母树叶瘿蚜均为蚊母新胸蚜(Neothoracaphis yanonis Matsumura),杭州新胸蚜(N.hangzhouensis)是蚊母新胸蚜的同物异名。 To identify the species of leaf gall-inducing aphids of Distylium racemosum in China and explore the population genetic differentiation,the COI and COII gene sequences of the leaf gall-inducing aphids of Distylium racemosum from Hunan,Chongqing,Jiangsu and Zhejiang were obtained by PCR amplification,and then were sequenced and compared with the homology sequences from GenBank.The phylogenetic trees based on COI and COII gene sequences of aphids were constructed by the NJ method.The results have shown that the samples from Nanjing,Changsha,Loudi and Yongzhou share the same unique haplotype.There is a certain genetic differentiation in the populations from Chongqing and Hangzhou,but the haplotype of the most samples is the same as that of Changsha and Nanjing populations,and the genetic distances among different haplotypes are extremely small.The results of sequence analyses reveal that all samples from six cities of four regions are Neothoracaphis yanonis Matsumura whose synonym is Neothoracaphis hangzhouensis Zhang.
