基于生信数据库研究ABCC5在肝细胞肝癌中的表达及其临床意义

摘要 目的 通过对肿瘤相关数据库的数据挖掘,研究ABCC5在肝细胞肝癌(LIHC)中的表达及临床意义。方法 使用TCGA数据库作ABCC5在LIHC中基因富集分析,使用GEPIA、LinkedOmics与GDSC数据库分析ABCC5与LIHC临床参数之间的关系。结果 ABCC5在LIHC癌组织中转录水平高于癌旁组织(P<0.01)。ABCC5表达水平与LIHC临床分期呈显著正相关(P<0.05),ABCC5高表达患者的术后总生存率(P<0.01)和无病生存率(P=0.03)均显著低于ABCC5低表达患者。ABCC5与LIHC的N分期(P<0.05)、肿瘤残余(P<0.05)呈显著正相关,而与T分期、M期、肿瘤放疗和年龄等因素无显著相关性(P均>0.05)。在LIHC细胞系中,ABCC5表达水平与多种化疗药物呈显著正相关(P均<0.05),基因富集分析结果显示:ABCC5与WNT和Hedgehog信号通路的激活有关(P均<0.05)。结论 ABCC5在LIHC组织中表达水平增高,ABCC5在LIHC组织中高表达意味着不良的临床分期、预后以及更强的耐药性,其分子机制可能与WNT和Hedgehog信号通路的激活有关。 Objective To study the expression and clinical significance of ABCC5 in hepatocellular carcinoma(LIHC) through tumor-related databases.Methods TCGA database was used for Gene Set Enrichment Analysis of ABCC5 in LIHC.The relationship between ABCC5 and LIHC clinical parameters was analyzed using GEPIA,LinkedOmics and GDSC databases.Results The expression level of ABCC5 in LIHC cancer tissues was higher than that in adjacent tissues(P<0.01).The expression level of ABCC5 was significantly positively correlated with the clinical stage of LIHC(P<0.05).The postoperative overall survival rate(P<0.01) and disease-free survival rate(P=0.03) of patients with high ABCC5 expression were significantly lower than those of patients with low ABCC5 expression.ABCC5 was significantly positively correlated with N stage(P<0.05) and tumor residual(P<0.05),but had no significant correlation with T stage, M stage, tumor radiotherapy and age(P>0.05) of patients with LIHC.In LIHC cell lines, ABCC5 were significantly correlated with multiple chemotherapeutics(P<0.05),and results of Gene Set Enrichment Analysis showed that ABCC5 was related to the activation of WNT and Hedgehog signaling pathways(P<0.05).Conclusion The expression of ABCC5 is increased in LIHC tissues.High expression of ABCC5 in LIHC tissues means poor clinical stage, prognosis, and stronger drug resistance.The molecular mechanism may be related to the activation of WNT and Hedgehog signaling pathways.
