基于数据挖掘和网络药理学探讨中医药治疗骨质疏松症的用药规律及机制 被引量:7

Regularity and Mechanism of Traditional Chinese Medicine in the Treatment of Osteoporosis Based on Data Mining and Network Pharmacology

摘要 目的:基于数据挖掘和网络药理学,探讨中医药治疗骨质疏松症的用药规律及机制。方法:应用中医传承辅助平台(V2.5),对2015—2022年中国知网收录的中医药治疗骨质疏松症的方剂进行用药规律挖掘。采用网络药理学方法,通过中药系统药理学数据库与分析平台检索化合物及治疗靶点,通过GeneCards等数据库筛选骨质疏松症的靶点并构建可视化蛋白质-蛋白质相互作用网络,对关键靶点进行基因本体(GO)功能富集分析及京都基因与基因组百科全书(KEGG)通路富集分析,构建成分-疾病-靶点网络图。结果:共纳入104张处方,使用频次居前3位的中药为熟地黄、淫羊藿及当归;使用频次最高的药对为“熟地黄→淫羊藿”,关联性最高的药对为“山药→熟地黄”;演化得到9个推荐的新方。共筛选出药物活性成分67个,药物作用靶点682个,疾病靶点2374个,交集靶点109个,GO功能富集分析得到氧化应激的反应、细胞对化学应激等1939个生物过程,KEGG通路富集分析得到晚期糖基化终末产物(AGE)-AGE受体、肿瘤坏死因子等159条信号通路。结论:治疗骨质疏松症的中药大多为补肝肾、强筋骨类药物,与骨质疏松症的基本病机相呼应,核心药物通过多成分、多靶点、多通路治疗疾病。 OBJECTIVE:To probe into the regularity and mechanism of traditional Chinese medicine in the treatment of osteoporosis based on data mining and network pharmacology.METHODS:The traditional Chinese medicine inheritance auxiliary platform(V2.5)was used to mine the medication rules of traditional Chinese medicine prescriptions for osteoporosis included in CNKI from 2015 to 2022.Network pharmacology method was used,TCMSP was adopted to search for compounds and therapeutic targets,GeneCards and other databases were used to screen osteoporosis targets and build the visual PPI network.Gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed on the key targets,component-disease-target network diagram was constructed.RESULTS:A total of 104 prescriptions were included,and the top 3 traditional Chinese medicines were Rehmannia glutinosa,Epimedium and Angelica sinensis;the drug pair with the highest frequency was“Rehmannia glutinosa→Epimedium”,and the drug pair with the highest correlation was“Yam→Rehmannia glutinosa”.Evolution obtained 9 recommended new recipes.A total of 67 active pharmaceutical ingredients,682 drug targets,2374 disease targets and 109 intersection targets were screened out.GO analysis obtained 1939 biological processes such as oxidative stress response and cell response to chemical stress.KEGG analysis obtained 159 signaling pathways including advanced glycation end products(AGE)-AGE receptor,tumor necrosis factor.CONCLUSIONS:Most of the traditional Chinese medicines for the treatment of osteoporosis are liver and kidney,muscle and bone strengthening drugs,which echo the basic pathogenesis of osteoporosis.Core drugs treat the disease through multi-component,multi-target and multi-pathway.
