烟草腺毛中NtHD-ZIP IV和NtJAZ家族基因表达分析 被引量:1

摘要 【目的】为探究NtHD-ZIP IV和NtJAZ家族基因在腺毛发生发育中的作用。【方法】通过实时定量PCR技术分析NtHD-ZIP IV和NtJAZ家族基因在高分泌型烤烟种质T.I.1068和非分泌型烤烟种质C110上的转录水平以及茉莉酮酸甲酯(Methyl Jasmonate,MeJA)处理C110后NtHD-ZIP IV和NtJAZ家族基因的转录水平。【结果】(1)C110的腺毛类型以短柄分泌型腺毛和非分泌型腺毛为主,T.I.1068的腺毛类型以长柄分泌型腺毛和短柄分泌型腺毛为主,MeJA处理后,C110的长柄分泌型腺毛密度增加。(2)Nt HD-ZIP IV-6、-7、-9和-14在C110和T.I.1068表皮毛中的表达水平均较高,NtHD-ZIP IV-1、-2、-3和-13在C110表皮毛中表达水平较高,NtHD-ZIP IV-12则在T.I.1068表皮毛中表达水平稍高。MeJA处理后,NtHD-ZIP IV-1、-2和-7的转录水平受到抑制,而NtHD-ZIP IV-5的转录水平上调。(3)与C110相比,Nt JAZ-1、-7、-9、-12、-13、-14和-15在T.I.1068表皮毛中的表达水平显著上调;与T.I.1068相比,Nt JAZ-11则在C110表皮毛中表达水平显著上调。MeJA处理后,NtJAZ-1、-3、-5、-7、-8、-9、-10、-12和-15的转录水平上调。【结论】MeJA对长柄分泌型腺毛有诱导作用,NtHD-ZIP IV-12和NtJAZ-1、-7、-9、-12、-13、-14、-15在T.I.1068表皮毛中的表达水平较高,而在C110表皮毛中表达水平较低。 [Background]This study aims to explore the role of NtHD-ZIP IV and NtJAZ family genes in glandular trichomes development.[Methods]The transcription levels of NtHD-ZIP IV and NtJAZ family genes in high-secreting flue-cured tobacco germplasm T.I.1068 and non-secreting flue-cured tobacco germplasm C110 as well as the transcription levels of NtHD-ZIP IV and NtJAZ family genes after MeJA treatment of C110 were analyzed by real-time quantitative PCR technology.[Results](1)The glandular trichomes types of C110 were mainly short-stalk secretory glandular trichomes and non-secreting glandular trichomes,while the glandular hair types of T.I.1068 were mainly long-stalk secretory glandular trichomes and short-stalk secretory glandular trichomes.The density of long-stalk secretory glandular trichomes of C110 increased after MeJA treatment.(2)The expression levels of NtHD-ZIP IV-6,-7,-9 and-14 were higher in C110 and T.I.1068 stem epidermis,while NtHD-ZIP IV-1,-2,-3 and-13 were higher in the stem epidermis of C110,and the expression level of NtHD-ZIP IV-12 was slightly higher in the stem epidermis of T.I.1068.After MeJA treatment,the transcriptional levels of NtHD-ZIP IV-1,-2 and-7 were inhibited,while that of NtHD-ZIP IV-5 was up-regulated.(3)Compared with C110,the expression levels of NtJAZ-1,-7,-9,-12,-13,-14 and-15 were higher in the stem epidermis of T.I.1068;compared with T.I.1068,the expression level of NtJAZ-11 was higher in the stem epidermis of C110.Transcript levels of NtJAZ-1,-3,-5,-7,-8,-9,-10,-12 and-15 were up-regulated after MeJA treatment.[Conclusion]MeJA can induce long-stalk secretory glandular trichomes,and the expression levels of NtHD-ZIP IV-12 and NtJAZ-1,-7,-9,-12,-13,-14,-15 were higher in T.I.1068 epidermal trichomes but lower in C110 epidermal trichomes.
