乳腺癌并发抑郁症模型小鼠血液及组织中白介素类细胞因子含量的变化

Changes of interleukin-like cytokines in blood and tissues of mice with breast cancer related with depression

摘要 目的:观察乳腺癌并发抑郁症(BCRD)模型小鼠行为、白介素类细胞因子的变化。方法:BALB/c雌性小鼠,腋下接种1×106个4T1炎症乳腺癌细胞,7 d后观察是否成模,成模后的小鼠连续21 d背部皮下注射皮质酮(30 mg/kg)复制BCRD模型。所有小鼠按糖水消耗随机分为4组:乳腺癌组、抑郁症组、BCRD组、空白对照组。21 d后,测定各组小鼠糖水偏嗜度、旷场实验等行为学变化;采用悬液芯片技术检测血清、海马、肿瘤、皮质组织中IL-1β、IL-2、IL-6、IL-10、IL-12含量。结果:与空白组比较,BCRD小鼠蔗糖偏嗜度及垂直活动次数降低(P<0.01或P<0.05),血清、海马中IL-2、IL-6、IL-12含量显著升高(P<0.01或P<0.05),海马中IL-10含量显著增加(P<0.01或P<0.05);与乳腺癌组比较,BCRD组血清、海马、皮质中IL-6含量显著升高(P<0.05),与抑郁症组比较,血清、海马、皮质中IL-6含量差异无统计学意义(P>0.05),但均有升高趋势。结论:IL-6在血清、海马、皮质中表达均较高且均一,可能是BCRD的重要标志物。 Objective:To observe the changes of behavior and interleukin-cytokines in mice with breast cancer related with depression(BCRD).Methods:The female BALB/c mice were inoculated with 1×1064T1 inflammatory breast cancer cells under the armpit.After 7 days,the mice were injected subcutaneously with corticosterone(30 mg/kg)in the back for 21 days to replicate the BCRD model.All mice were randomly divided into four groups according to the consumption sugar water:breast cancer group,depression group,BCRD group,and normal group.After 21 days,the behavioral changes of mice in each group,were measured.such as sugar water preference and open field experiment,The contents of IL-1β,IL-2,IL-6,IL-10 and IL-12 in serum,hippocampus,tumor and cortex were measured by suspension chip technology.Results:Compared with normal group,the sucrose preference degree,horizontal and vertical activity times of BCRD mice decreased(P<0.01 or P<0.05),the contents of IL-2,IL-6 and IL-12 in serum,hippocampus and cortex were increased significantly(P<0.01 or P<0.05),and the contents of IL-10 in hippocampus was increased significantly(P<0.01 or P<0.05).Compared with breast cancer group,the IL-6 content in serum,hippocampus and cortex in the BCRD group was significantly increased(P<0.05),and compared with the depression group,there was no significant difference in the IL-6 content in serum,hippocampus and cortex(P>0.05),but all showed an increasing trend.Conclusion:The expression of IL-6 in serum,tumor,hippocampus and cortex is high and uniform,which may be an important marker of BCRD.
