纳晶微针负载米诺地尔治疗小鼠雄激素性脱发的疗效

Effect of nanocrystalline microneedles loaded with minoxidil in the treatment of androgenetic alopecia in mice

摘要 目的:探究负载米诺地尔的纳晶微针对雄激素性脱发的C57BL/6小鼠毛囊及毛发生长的影响。方法:C57BL/6小鼠背部皮下注射丙酸睾酮诱导雄激素性脱发动物模型,造模成功后进行随机分组,各组按要求给予不同处理,观察并记录小鼠毛发变化,然后在第10周取各组小鼠背部皮损,作病理切片行HE染色测量毛囊数量及终毛样毛囊/毳毛样毛囊比例。结果:负载米诺地尔的纳晶微针毛发生长评分明显大于造模阴性组,其毛囊数量以及终毛样毛囊/毳毛样毛囊比例均显著高于造模阴性组(P<0.05),且不低于米诺地尔组。值得注意的是,空白微针组也有一定的促进毛发生长的作用。结论:负载米诺地尔的纳晶微针可以有效地将米诺地尔缓慢、直接、持续地运输到毛囊周围,并在减少米诺地尔剂量和治疗次数的情况下,达到较好的治疗效果。这将为其临床应用奠定一定基础。 Objective:To explore the effect of minoxidil⁃loaded nanocrystalline microneedles on hair folli⁃cles and hair growth in C57BL/6 mice with androgenetic alopecia.Methods:An animal model of an⁃drogenetic alopecia induced by subcutaneous injection of testosterone propionate on the back of C57BL/6 mice.After successful modeling,the mice were randomly divided into groups,and each group was given different treatments as required,and the hair changes of mice were observed and re⁃corded.At the end of the 10th week post⁃treatment,the skin samples of mouse's back in each group were taken,and the pathological sections were taken for HE staining to measure the number of hair follicles and the ratio of terminal hair follicles/vellus hair follicles.Results:The hair growth score of the nanocrystalline microneedle loaded with minoxidil was significantly higher than that in the negative model group,and the number of hair follicles and the ratio of terminal hair follicles/vellus hair follicles were significantly higher than those in the negative model group(P<0.05),and no lower than the top⁃ically applied minoxidil group.It is worth noting that the blank nanocrystalline microneedle group also has a certain effect on promoting hair growth.Conclusion:Nanocrystalline microneedles loaded with minoxidil can effectively transport minoxidil to the hair follicles slowly,directly and continuously,and achieve good therapeutic effects under the condition of reducing the dose of minoxidil and the times of treatments,which will lay a solid foundation for clinical application.
