摘要
为了解我国禽类养殖场中Ⅰ群禽腺病毒(FAdV-Ⅰ)的分子流行病学情况,对采自6个不同省份及地区的324份疑似FAdV感染组织利用PCR进行病原学检测,通过扩增其Hexon基因并连接至pMD19-T载体进行测序和遗传进化分析。结果显示,324份样品中阳性检出率为23.4%,鸡、鹅、鹌鹑、鸽、鸭中均有检出,其中2021年FAdV的检出率高达61.8%。另外,在扩繁得到76株病毒中,通过序列对比发现,血清1型(FAdV-1)占比较少为7.9%,血清4型(FAdV-4)占比较高为92.1%。FAdV-1样本株与国内外流行毒株处于同一遗传进化分支,Hexon L1区域的核苷酸同源性为98.7%~100%,部分FAdV-4样本株与中国参考毒株处于同一遗传进化分支。本研究在2015—2022年采集了我国部分地区疑似FAdV感染的病料进行Ⅰ群禽腺病毒的分子流行病学调查和遗传进化分析,为进一步做好相关防控工作提供理论参考。
In order to understand the molecular epidemiology of fowl adenovirus groupⅠ(FAdV-1)in poultry farms in China,324 suspected FAdV-infected tissues collected from six different provinces and regions were detected by PCR,and their Hexon genes were amplified and connected to pMD19-T vector for sequencing and genetic evolution analysis.The results showed that the positive detection rate of 324 samples was 23.4%,and all species of chickens,geese,quails,pigeons and ducks were detected,among which the detection rate of FAdV was as high as 61.8%in 2021.In addition,among 76 virus strains obtained by propagation,it was found through sequence comparison that,FAdV-1 accounted for 7.9%and FAdV-4 accounted for 92.1%.The FAdV-1 sample strain is in the same genetic evolution branch with the domestic and foreign epidemic strains,and the nucleotide homology of Hexon L1 area is 98.7%—100%,while some FAdV-4 sample strains are in the same genetic evolution branch with the Chinese reference strains.From 2015 to 2022,this study collected samples of suspected FAdV infection in some areas of China,and carried out molecular epidemiological investigation and genetic evolution analysis of fowl adenovirus groupⅠ,so as to provide theoretical reference for further prevention and control.
